摘要
目的探讨β-elemene联合顺铂对体外培养的Hep-2细胞(人喉咽癌细胞)生长的抑制作用。方法采用MTT比色法和流式细胞分析技术检测不同组别、不同时间(12、24、48h)对Hep-2细胞的抑制其增殖和诱导其凋亡的作用。结果单纯β-elemene组、单纯顺铂组及β-elemene联合顺铂组,12h时对Hep-2细胞增殖抑制率分别为19.05%、20.12%和22.24%,细胞凋亡率分别为12.21%、17.98%和20.34%;24h时细胞增殖抑制率为40.62%、41.51%和64.65%,细胞凋亡率为25.34%、31.08%和34.62%;48h时细胞增殖抑制率为60.50%、68.23%和71.56%,细胞凋亡率为40.63%、42.96%和63.01%。β-elemene联合顺铂组对Hep-2细胞生长抑制作用和诱导细胞凋亡的作用都明显优于其他各组(P<0.05)。结论β-elemene联合顺铂能够明显抑制Hep-2细胞增殖,诱导Hep-2细胞凋亡,为喉咽癌的化疗提供了一个新的启发。
Objective To investigate the effect of the growth inhibition of β-elemene combined with eisplatin on the cultured Hep-2 cells (human hypopharyngeal carcinoma cells) . Methods MTT and Flow cytometry was applied to detect the inhibition and the apoptosis of human Hep-2 cells on different groups at different times (12 h, 24 h, 48 h) . Results The 12 h cells proliferation inhibition rates were 19.05%, 20.12% and 22.24% in elemene group, cisplatin group and elemene combined with cisplatin group, and the apoptosis rates were 12.21 %, 17.98% and 20. 34% ; The 24 h cells proliferation inhibition rates were 40.62%, 41.51% and 64.65%, and the apoptosis rates were 25.34%, 31.08% and 34.62% ; The 48 h cells proliferation inhibition rates were 60.50 %, 68.. 23 % and 71.56 %, and the apoptosis rates were 40.63 %, 42.96 % and 63.01 %. The growth inhibition and induction of apoptosis on Hep-2 cell of β-elemene combined with cispiatln group were significantly better than the other groups (P d0.05) . Conclusion β-elemene combined with cisplatin can inhibit the proliferation of Hep-2 ceils, induce apoptosis of Hep-2 cells, which can provide a new inspiration for the hypopharyngeal carcinoma chemotherapy.
