SD大鼠两种移植性胃癌模型的诱癌成功率比较

Comparison of success rates among two different situ human gastric tumor metastasis model in SD rats

目的用瘤细胞悬液和瘤组织块原位种植,建立SD大鼠原位移植胃癌模型。方法直接注射法组将walker-256细胞悬液直接注射到SD大鼠胃壁的浆膜内,观察移植肿瘤的生长状况、移植成功率;组织插块法组以walker-256细胞接种于SD幼鼠皮下,形成稳定传代的皮下移植瘤,再取该肿瘤组织块原位移植于SD大鼠胃壁,观察比较移植肿瘤的生长状况、移植成功率。结果直接注射组原位移植成功率为71.42%,造模九天后平均瘤重为0.322 g;组织插块组原位移植胃癌成功率为42.85%,造模九天后平均瘤重为0.206 g。两组间比较有统计学意义(P<0.01)。结论该SD大鼠直接注射法原位移植模型的生物学行为与人胃癌自然生长程相似,可作为一种有价值的工具用于胃癌转移机理和抗转移实验。

Objective Tumor tissue and tumor cell suspension was inoculated into SD rats to establish orthotopic gastric cancer model.Methods Walker-256 cells were inoculated subcutaneously into SD rats ,then intact tumor tissue was collected and implanted orthotopically into SD rat stomach , the model was named indrect gastric orthotopic transplantation .To observe the tumor growth condi-tions and the success rate of transplantation .Walker-256 cell suspension will be directly injected into the stomach wall serous membrane of SD rats in direct injecrion group ,tumor growth conditions and the success rate of transplantation were observed .Results Situ trans-plant success rate in direct group was 71.42%, nine days after modeling , the average tumor weight was 0.322 g;situ tissue transplant success rate of indirect gastric orthotopic transplantation group was 42.85%,nine days after modeling , the average tumor weight was 0. 206 g.Conclusion Direct injection of the SD rat model of orthotopic transplantation is similar to human gastric cancer biological be -havior and natural growth process ,it can be used as a valuable tool for the transfer mechanism of gastric cancer and anti -transfer ex-periments .
