胃癌中SOX18、VEGF-C和VEGFR-3的表达及意义 被引量:5

摘要 目的探讨SOX18、血管内皮生长因子C(vascular endothelial growth factor-C,VEGF-C)和血管内皮生长因子受体3(vascular endothelial growth factor receptor-3,VEGFR-3)在胃癌中的作用。方法利用免疫组化SP法检测60例胃癌及15例对应远端正常胃黏膜组织中SOX18、VEGF-C和VEGFR-3的表达。结果胃癌中SOX18、VEGF-C和VEGFR-3的阳性率显著高于正常黏膜组织,差异具有统计学意义(P<0.05)。胃癌中SOX18、VEGF-C蛋白表达与肿瘤浸润深度、淋巴结转移及TNM分期有关(P<0.05),VEGFR-3蛋白表达与肿瘤淋巴结转移及TNM分期有关(P<0.05)。胃癌组织中SOX18与VEGF-C(r=0.372,P=0.002)、VEGFR-3(r=0.308,P=0.011),VEGF-C与VEGFR-3(r=0.333,P=0.005)阳性表达均存在显著正相关。结论 SOX18、VEGF-C和VEGFR-3可能在胃癌的发生、发展和淋巴结转移中发挥重要作用。SOX18可能通过调控VEGF-C表达介导淋巴管生成,从而促进胃癌的淋巴结转移。 Purpose To investigate the roles of SOX18, vascular endothelial growth factor-C (VEGF-C) and its receptor VEGFR-3 in gastric carcinoma. Methods The expression of SOXI8, VEGF-C and VEGFR-3 was detected by SP immunohistochemical technique in 60 cases of gastric carcinoma and their corresponding adjacent normal gastric tissues ( n = 15 ). Results The positive percentage of SOX18, VEGF-C and YEGFR-3 protein expression in gastric cancer was significantly higher than that in normal gastric mucosa tissues (P 〈0. 05). The expression of SOX18 and VEGF-C was significantly related with invasive depth, lymph node metastasis and TNM stage ( P 〈 0.05 ), the expression of VEGFR-3 was significantly related with lymph node metastasis and TNM stage ( P 〈 0.05 ). The expression of SOX18 had significantly positive correlations with that of VEGF-C (r =0. 372, P =0. 002) and VEGFR-3 (r =0. 308, P = 0. 011 ) in gastric carcinoma, and there was also significantly positive correlation between expression of VEGF-C and that of VEGFR-3 in gastric carcinoma (r = 0. 333, P = 0. 005). Conclusions SOX18, VEGF-C and VEGFR-3 may play an important role in the pathogenesis, development and lymph node metastasis of gastric carcinoma. SOX18 may mediate lymphangiogenesis through regulating the expression of VEGF-C, thereby promoting the lymph node metastasis of gastric carcinoma.
