凹顶藻萜类化合物对酒精暴露大鼠肝脏bcl-2和bax表达的影响

摘要 目的了解凹顶藻萜类化合物(LTE)对酒精暴露大鼠肝脏超微结构及凋亡相关基因bcl-2和bax表达水平的影响。方法 Wistar大鼠随机分组,每日予50%酒精4.8 g·kg-1·d-1灌胃,建立酒精暴露模型;试验组每日分别给予LTE 25、50、100 mg/kg灌胃给药;正常对照组给予等体积蒸馏水。应用透射电镜观察大鼠肝细胞超微结构的变化,免疫组化染色法检测bcl-2和bax表达水平及bcl-2/bax值。结果酒精模型组大鼠肝细胞脂肪变形,电镜下观察到轻度炎性细胞浸润,线粒体肿胀变形,粗面内质网扩张断裂,试验组的肝细胞破坏程度均有不同程度减轻,bcl-2表达水平显著升高,bax表达水平显著降低,bcl-2/bax比值升高,差异有统计学意义(P<0.05)。结论一定剂量LTE可明显改善酒精暴露大鼠肝细胞超微结构病理变化,并使bcl-2/bax上调,从而减轻酒精性肝损伤。 Objective To study the effect of Laurencia terpenoid extract (LTE) on the hepatic ultra-structures and expression of apoptosis genes of rats in vivo after alcohol exposure. Methods Male Wistar rats assigned randomly to groups were orally given with 50% alcohol of 4.8 g/kg everyday to build alcohol exposure model; test groups were orally fed with LTE of 25, 50 and 100 mg/kg everyday respectively, normal control group was given with distilled water. The hepatic ultra-structures of rats were studied by transmission electron microscope. The expression levels of bcl-2 and bax in hepatic tissue were detected by immunohistoehemistry method. Results The hepatic ultra-structures result showed that hepatocytes in alcohol model group steatosis, slight inflammatory cell infiltration, mitoehondria swollen deformative and dilated rough endoplasmlc fraetue. Test groups showed slight destroy degree. Compared with alcohol model group, the expression level of bel-2 in protective LTE groups and positive control group was increased significantly ( P 〈 0.05 ) , and the expression level of bax was decreased significantly (P 〈 0.05). Conclusion LTE at certain dosages can pro- mote mitochondria hyperplasy to improve hepatic ultra-structural pathological change, and raise the ratios of bel-2 and bax in hepatic tissues after alcohol exposure. So, LTE can effectively reduce the degree of injury of hepatic tissues by al- eohol .
