内源性上调pp-GalNAc-T4基因表达抑制全反式维甲酸诱导NB4白血病细胞分化的机制研究

摘要 目的上调pp-GalNAc-T4基因表达抑制全反式维甲酸诱导的NB4白血病细胞分化的机制探讨。方法用全反式维甲酸处理NB4白血病细胞和转染了pp-GalNAc-T4基因的NB4细胞及转染了空载体的NB4细胞,用实时PCR观察这三组细胞的PML-RARα基因表达情况。结果用全反式维甲酸处理转染了pp-GalNAc-T4基因的NB4细胞(NB4-T4细胞株)后,癌蛋白PML-RARα基因表达在NB4-T4细胞株,明显高于未转染pp-GalNAc-T4基因的NB4细胞和转染空载体的NB4细胞(P<0.01)。结论内源性上调pp-GalNAc-T4后,接着用全反式维甲酸诱导NB4-T4细胞。NB4-T4细胞中PML-RARα基因表达升高,而且NB4-T4细胞没有分化。这些提示,pp-GalNAc-T4可以抑制全反式维甲酸诱导的NB4细胞的分化作用,可能和癌蛋白PML-RARα有关。
