摘要
[目的]探讨大鼠前列腺去神经后组织中表皮生长因子(EGF)的变化及其对前列腺增生的影响。[方法]将130只成年SD雄性大鼠随机分成四组,对照组10只,实验组120只(实验组分为三组,每组40只),用0.01%戊巴比妥钠(1 mL/kg )腹腔注射麻醉,实验组切断支配前列腺的神经后缝合饲养,术后1周,2周,3周各处死40只,完整取出前列腺,分别用于电镜观察与免疫组化检测。[结果]去神经组前列腺体积缩小,组织水肿轻,无炎细胞浸润,HE染色与电镜下观察神经纤维显著减少,甚至消失,平滑肌逐渐变薄,腺细胞空泡变性,部分细胞器破坏,基底膜萎缩,胶原纤维轻度增生;EG F在各组均有表达,对照组呈强阳性,去神经组随时间延长,表达逐渐减弱,呈阳性、弱阳性、阴性趋势变化,术后第3周尤其明显;对照组与术后1周,术后1周与2周比较EGF表达差异无显著性(P>0.05)。对照组与术后2,3周比较,术后1,2周分别与3周比较差异均有显著性( P<0.05)。[结论]大鼠前列腺去神经后其体积缩小,腺组织中EG F的表达随时间的延长而减弱。
[Objective] To explore the influence of the changes of epidermal growth factor (EGF) in tissues after denervation of prostate in rats .[Methods] Totally 130 adult male SD rats were randomly divided into 4 groups .The control group had 10 rats ,while the experimental control group had 120 rats .All rats received the anesthesia by in-jecting 0 .01% sodium pentobarbital(1 mL/kg) .The experimental group was sutured and fed after dissecting the nerve of prostate .Every 40 rats were sacrificed 1 ,2 and 3 weeks after operation ,respectively .Prostates were com-pletely drawn for electron microscope examination and immunohistochemical analysis .[Results]In denervation group , the size of prostate was reduced ,and tissue edema was slight ,and no inflammatory cell infiltration was found .HE staining and electron microscope showed that nerve fibers were markedly decreased and even disappeared ,and smooth muscle became thinner ,and the glandular cells had vacuolar degeneration ,and some organelles were destroyed ,and the basement membrane became atrophy ,and collagen fibers had mild hyperplasia .EGF was expressed in each group . The expression of EGF in control group was strongly positive ,while that in denervation group was gradually de-creased with the prolongation of time ,and had the change trend of positive ,weakly positive to negative expression , and the change was more obvious 3 months after operation .There was no significant difference in EGF expression be-tween control group and experimental group 1 week after operation or experimental group 1 week and 2 weeks after operation( P>0 .05) ,but there was significant difference between control group and the experimental group 2 and 3 weeks after operation or the experimental group 1 or 2 weeks after operation and 3 weeks after operation( P<0 .05) .[Conclusion] The size of rat prostate is reduced after denervation .The expression of EGF in prostate tissue is de-creased with prolongation of time .
