摘要
[目的]研究喘可治口服液的抗过敏作用。[方法]观察不同剂量喘可治口服液对磷酸组胺所致豚鼠皮肤过敏反应的影响,计算过敏反应的致痒阈值。用2,4-二硝基氯苯(2,4-DNCB )对昆明小鼠皮肤致敏和诱发制成迟发型超敏反应模型。将小鼠随机分为5组,即模型对照组、阳性对照组、高剂量组、中剂量组、低剂量组,连续灌胃10 d ,d9对小鼠右耳激发过敏。观察记录激发后各组小鼠左右耳耳郭肿胀度、肿胀率、抑制率、胸腺指数及脾脏指数。利用卵白蛋白做抗原,百日咳疫苗做佐剂,制取大鼠抗过敏血清。同种雄性大鼠随机分为5组,即模型对照组、阳性对照组、高剂量组、中剂量组、低剂量组。将抗血清皮内注射于各组大鼠背部皮肤,利用抗原攻击,同时尾静脉注射1%伊文思蓝溶液,剪下带蓝斑皮肤于丙酮-生理盐水(7∶3)溶液,检测吸光度值。[结果]高剂量能明显提高组胺所致豚鼠致痒阈值,与模型对照组相比阈值差异有显著性。与模型对照组比较,高、中、低三剂量组小鼠耳郭的肿胀度、肿胀率、抑制率以及胸腺指数和脾脏指数均有所下降。高剂量组、中剂量组均能明显地抑制同种大鼠的被动皮肤过敏反应。[结论]喘可治口服液具有一定抗过敏及抗迟发型超敏反应作用,其中高剂量组作用较显著。
[Objective]To explore the anti-allergic effect of Chuankezhi oral solution .[Methods] The influence of different dose of Chuankezhi oral solution on skin allergic reaction in rats caused by histamine phosphate . Itch threshold of allergic reaction was calculated .The Kunming mouse model with the delayed hypersensitivity was estab-lished by skin allergization and provocation with 2 ,4-dinitrochlorobenzene(2 ,4-DNCB) .All mice were randomly di-vided into 5 groups including model control group ,positive control group ,high dose group ,middle dose group and low dose group .Mice received continuous lavage for 10 days .Right ear of mice received allergic stimulation at day 7 . The auricle swelling degree of left and right ear ,swelling rate ,inhibition rate ,thymus index and spleen index in all groups were observed and recorded after provocation .The eggalbumin and pertussis vaccine were used as antigen and adjuvant respectively to prepare rat anti-allergic serum .Homogenous male rats were randomly divided into 5 groups including model control group ,positive control group ,high dose group ,middle dosage group and low dosage group . The serum was intracutaneously injected into back skin of rats .Rats were challenged with antigen .Meanwhile ,vena caudalis was injected 1% evans blue followed by obtaining the skin with blue dot .The skin was then soaked in ace-tone/saline solution(7∶3) to test the absorbance value .[Results]Compared with model control group ,itch thresh-old of guinea pig caused by histamine in high dose group was increased obviously ,and there was significant difference between two groups .Compared with model control group ,auricular swelling degree ,swelling ratio ,inhibition ratio , thymus index and spleen index in high dose group ,middle dose group and low dose group were decreased .High dose and middle dose could obviously inhibit the passive dermal allergy in homogenous rats .[Conclusion]Chuankezhi oral solution has anti-allergic and delayed hypersensitive effect .Moreover ,the effect in high dose group is more signifi-cant .
