摘要
【目的】对三大道地产区的金银花品种进行遗传多样性分析,为道地产区种质资源利用与开发提供理论依据。【方法】应用ISSR分子标记技术对27个金银花原植物种质资源进行遗传多态性、相似性和聚类分析。【结果】筛选出16个多态性引物,获得344条带,多态性比率为88.4%;遗传相似系数在0.50~0.90之间;聚类分析表明,27个品种被分为2个类群。【结论IISSR标记可以应用于金银花样品间遗传多样性分析;道地产区的金银花种内不同地域或不同植物来源的材料间存在较大遗传差异。
[Objective] The objective of this study was to detect the genetic diversity of L. japonica from three authentic regions and to provide the theoretical basis for development and utilization of germplasm resources. [Method] Intersimple sequence repeat (ISSR) was used to analyze genetic polymorphism, similarity and cluster map of 27 honeysuckle germplasm resources. [Results] 344 bands were obtained from 16 selected primers. The percentage of polymorphic bands was 88.4%. The genetic similarity varied from 0.50 to 0.90. 27 varieties were divided into 2 groups by cluster analysis. [Conclusion] ISSR markers can be applied for genetic diversity of L. japonica. There were obvious genetic differences among regions and plant sources.
