超声波法虎杖中总黄酮提取工艺研究 被引量:7

摘要 目的确定超声波辅助方法提取虎杖中总黄酮的最佳工艺。方法采用正交实验优化影响虎杖中总黄酮类成分提取的4种主要因素:提取剂(CH3CH2OH)浓度、提取温度、超声时间以及料液比;并以芦丁为标准样品,利用紫外可见分光光度法测定虎杖中总黄酮的吸光度值,为确定最佳提取条件提供参考。结果通过正交实验得到从虎杖中提取总黄酮的最佳工艺为:乙醇浓度为75%,超声温度为50℃,超声时间为35min,料液比1∶40,其提取率可以达到7.68%。结论通过该实验确立了虎杖中总黄酮提取的最优工艺,提取效果良好、操作简便易行,节能环保。 Objective--To determine the optimum process of extracting total flavonoids from Polygonum cuspidatum by using the ultrasonic extraction technology. Methods--The orthogonal design experiment is used to optimize and influence the four dominant factors (the content of extraction agent (CH3 CH2 OH), extraction temperature, ultrasonic time and solidqiquid ratio) of extracting total flavonoids. Then rutin was as a reference object, the ultraviolet-visible spectrophotometry is uti- lized to determine the absorbance value of total flavonoids in Polygonum cuspidatum to provide the reference for optimal extraction parameters. Results --The research results show that the extraction efficiency of total flavonoids is 7.68%, when the ethanol concentration is 75%, the ultrasonic time is 35 min, the temperature is 50℃ and the solid-liquid ratio is one to forty. Conclusion--Through the experiment, not only are the optimal extraction conditions established but also the extraction effect is terrific. Moreover, the experiment is simple and easy as well as energy-saving and environment-friendly.
