慢病毒介导的Jab1基因表达干扰在乳腺癌细胞中功能的初步研究 被引量:1

摘要 在乳腺癌细胞MDA231中,成功建立了慢病毒介导的Jab1基因表达干扰系统,并通过MTT细胞增殖实验和体外细胞侵袭能力实验证实Jab1基因表达的干扰使得MDA231细胞的增殖和体外侵袭能力发生显著的下降;此外,通过Western blot实验证实Jab1对于Fra1的表达可能存在调控,并且Fra1和Jab1在MDA231细胞中的表达干扰会使细胞形态发生相似的改变,提示Jab1和Fra1在乳腺癌细胞MDA231中可能通过调控共同的信号通路影响细胞的形态. With lenti-virus mediated RNA interference, a Jab1 gene knockdown study system in breast cancer cell line MDA231 was established. By using MTT cell proliferation assay and cell invasion assay, we confirmed that the knockdown of gene Jab1 expression could result in a significant atienuation of the proliferation and in vitro invasion capability of MDA231, indicating the critical role of Jab1 in the proliferation and invasion of breast cancer cells. Besides, Western Blot experiment result showed that Jabl could regulate the expression of Fral, whose expression interference in MDA231 cell could cause a simliar morphologic change of the cell as Jabl, suggests that Jabl and Fral may have a co-effect on the signal transduction with regard to the cellular morphology.
