摘要
目的探讨丹参注射液抑制肺纤维化的作用及其机制。方法将40只健康雌性SD大鼠采用随机数字表法分为正常组、丹参组、地米组、模型组,每组10只,丹参组、地米组、模型组采用气管内注入博莱霉素建立SD大鼠肺纤维化模型。观察大鼠肺系数、肺纤维化评分、肺组织病理形态学改变等,并通过实时定量聚合酶链反应(RT-PCR)检测大鼠肺组织转化生长因子-β1(TGF-β1)及基质金属蛋白酶-9(MMP-9)mRNA的表达。结果丹参组及地米组肺系数明显低于模型组,但高于正常组,差异均具有统计学意义(P<0.01),同时丹参组低于地米组(P<0.01);肺组织病变呈肺纤维化改变,与模型组比较,_丹参组及地米组明显较轻(P<0.01),且丹参组优于地米组,差异具有统计学意义(P<0.01)。RT-PCR检测结果提示模型组大鼠TGF-β1及MMP-9明显高于丹参组及地米组(P<0.01),丹参组也明显低于地米组(P<0.01)。结论丹参注射液能够减轻肺纤维化,其作用机制可能与下调TGF-β1及MMP-9 mRNA的表达有关。
Objective To observe the effect of Danshen injection on inhibition of pulmonary fibrosis and its mechanism. Methods Forty healthy SD rats were divided into normal group,Danshen(DS) group,dexamethasone(DEX) group and model group (n = 10/group). Bleomycin was used to induce pulmonary fibrosis in all the rats except those in the normal group. Lung coefficien, fi- brosis score and pathological changes were explored. Real-time fluorescent quantitative polymerase chain reaction was used to probe the expression of transforming growth factor -β1 (TGF-β1) and matrix metalloproteinase-9 ( MMP-9). Results After intratracheal infusion of bleomycin, the lung coefficien, the fibrosis score and the expression of TGF-β1 and MMP-9 in the DS and DEX group were remarkably lower than that of the model group, and the above indictors in the DS group were significantly lower than that of the DEX group ( P 〈 0.01 ). It was showed that inflammatory reaction was obvious in the model group, and its fibrosis degree was se- vere, while the changes were significantly attenuated in the DS group and the DEX group ( P 〈 0.01 ). All test data were the lowest in the normal group, and there was no obvious pulmonary fibrosis in the normal group. Conclusion Danshen injection shows an- tagonism against bleomycin induced pulmonary fibrosis probably related to inhibition of TGF-β1 and MMP-9 mRNA transcription.
