凝血酶敏感蛋白1对体外培养大鼠肺动脉平滑肌细胞的影响

摘要 目的了解凝血酶敏感蛋白1(TSP-1)对体外培养的大鼠肺动脉平滑肌细胞(PASMCs)的影响。方法体外培养大鼠PASMCs,加入不同浓度(10-12、10-11和10-10 mol/L)TSP-1处理12、24、48h,采用3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐(MTT)法检测细胞增殖,流式细胞术(FCM)检测细胞周期。结果 TSP-1刺激后,MMT法实验结果显示TSP-1显著促进大鼠PASMCs增殖,此作用呈浓度和时间依赖性。FCM分析显示TSP-1使S期细胞比例增加。TSP-1处理12h后,S期细胞比例增加,24h时轻微下降,48h时达高峰。结论 TSP-1呈浓度和时间依赖性地促进大鼠PASMCs增殖。 Objective To investigate the effect of thrombospondin 1(TSP-1)on the cell proliferation of cultured rat pulmonary artery smooth cell(PASMCs)in vitro.Methods Rat PASMCs were cultured in vitro,and treated with different concentrations(10^-12、10^-11、10^-10 mol/L)of TSP-1for 12,24,48h.The cell proliferation was quantified by MTT assay.The cell cycle of PASMCs was measured by flow cytometric(FCM)analysis.Results MTT assay showed that TSP-1promoted the proliferation of PASMCs significantly,and the effect was concentration-dependent and time-dependent.FCM analysis indicated that TSP-1increased the percentage of S phase.The percentage of S phase of PASMCs were increased after treated with thrombospondin-1for 12h, slight down after 24h,while reached a maximal level at 48h.Conclusion The TSP-1promotes rat PASMCs proliferation in a concentration-dependent and time-dependent manner.
