油酸对鸡前脂肪细胞分化过程中基因表达的影响 被引量:7

摘要 采用酶消化原代培养脂肪细胞的方法培养肉鸡前体脂肪细胞,在培养基中加入1 mmol·L-1油酸后,分别在24,48,72和96 h检测细胞中PPARγ基因,SREBP1基因、ApoAI基因、A-FABP基因、IGF-I基因和IGFIR基因的表达变化,同时对细胞中葡萄糖及甘油三酯的含量进行检测。结果表明,加入油酸48 h后前脂肪细胞中开始出现脂滴。油酸诱导后24 h的前脂肪细胞中,处理组中脂类代谢相关基因ApoAI基因、A-FABP基因的表达量显著高于对照组,处理组中参与脂质合成的甘油三酯和葡萄糖的含量随着时间不断积累。结果显示,油酸通过诱导脂类代谢基因ApoAI基因,A-FABP基因的表达进而使脂滴在前脂肪细胞中沉积。 In this study, the chicken preadipocyte were cultured by the method of enzyme digestion. After added oleate into the medium of cultured cells, the expression level of PPARy, SREBP1, ApoAI, A-FABP, IGF-I, IGF-IR were detected at 24, 48, 72 and 96 h by real-time PCR. The content of tdglyedde and glucose in adipocyte were also detected. The result showed that the lipid droplets begin appearing in adipocytes at 48 h after the oleate added. The expression level of ApoAI and A-FABP in adipocyte were upregulated at 24 h after treated with oleate. The content of triglyeride and glucose in adipocyte were increased during the time that treated with oleate. These results suggested that oleate could induce differentiation of chicken preadipocytes through upregulating the expression of ApoAI and A-FABP gene.
