抑癌基因beclin1在子宫内膜异位症及非子宫内膜异位症患者中差异表达的意义

Differential expression of the anti-oncogene beclin 1 between patients with and without endometriosis

目的研究子宫内膜异位症(cndometriosis,Ems)组织中抑癌基因beclin 1的mRNA和蛋白表达情况。方法利用RT-PCR和Western Blot免疫印迹的方法,对EMs的异位内膜组织、在位内膜组织和非EMs在位内膜组织(正常内膜组织)进行mRNA和蛋白表达的半定量分析。利用原位杂交和免疫组化技术对研究各组进行定性及定位分析。结果①beclin 1 mRNA和蛋白在各组中均有表达,异位内膜的表达高于在位内膜和正常内膜,差异有显著性(P<0.005),在位内膜组与正常对照组比较,beclin 1的表达无明显差异(P>0.05);②原位杂交阳性率分别为非EMs组95.1%,EMs异位内膜组100%,EMs在位内膜组91.4%;③免疫组化阳性率分别为非EMs组91.7%,EMs异位内膜组100%,EMs在位内膜组93.4%;④beclin 1的DNA与蛋白表达一致;⑤7例恶变的异位内膜beclin 1免疫组化检测,5例呈阴性,2例呈弱阳性表达。结论抑癌基因beclin 1在子宫内膜组织中均表达,且在异位内膜中呈表达上调,而在恶变的异位内膜中多呈阴性表达,提示beclin 1基因可能与子宫内膜异位症不孕及恶变相关。

【Objective】To investigate the expression of the anti-oncogene beclin 1 in the endometriotic tissue and explore its clinical significance. 【Methods】A semiquantitative analysis of the expression of beclin 1 mRNA and protein in the ectopic and eutopic endometrium of women with endometriosis was conducted using RT-PCR and Western blotting in comparison with that in the endometrium of women without endometriosis. Immunohistochemisty and in situ hybridization was performed for semiqualitative analysis and localization of ARHI expression.【Results】 The expression levels of beclin 1 mRNA and protein differed significantly between the groups. Beclin 1 was ex- pressed at significantly higher levels in ectopic endometrium than in eutopic and normal endometrium（P 0.05）, butwhichshowed no significant difference between the latter two tissues（P 0.05）. The positivity rate of beclin 1 DNA was 95.1% in the endometrium of women without endometriosis, 100% in the ectopic endometrium and 91.4% in the eutopic endometrium of women with endometriosis. Imrnunohistochemisty showed an ARHI positivity of 91.7% in the endometrium of women without endometriosis, 100% in the ectopic endometrium and 93.4% in the eutopic en- dometrium of women with endometriosis. The expression patterns of beclin 1 DNA and protein were consistent. Im- munohistochemisty in 7 cases of malignant endometriosis showed negative ARHI expression in 5 cases and weak positivity in 2 case.【Conclusion】Beclin 1 expressions are present in the endometrium and up-regulated in ectopic endometrium, whereas in the ectopic endometriam of patients with malignant endometriosis its expression is often negative, suggesting a role of beclin 1in infertility and tumorigenesis of endometriosis.
