化香树果序挥发油的气相色谱-质谱联用分析及体外抗肿瘤活性研究 被引量:9

GC-MS Analysis and Antitumor Activity in Vitro of Chemical Constituents of Essential Oil from Platycarya Strobilacea

摘要 目的:研究化香树果序挥发油的化学成分及其体外抗肿瘤活性。方法:采用水蒸气蒸馏法从化香树果序中提取挥发油,通过气相色谱-质谱联用技术对其挥发油成分进行分析;采用MTT法观察其对人肝癌HepG2细胞、人鼻咽癌CNE-2细胞及人宫颈癌HeLa细胞的增殖抑制率及半数抑制浓度(IC50)。结果:从挥发油中共鉴定出28个化合物,占总离子流图峰面积的94.10%,其中含量较高的组分为(Z,Z,Z)-9,12,15-十八碳三烯酸(44.48%)、十六烷酸(23.30%)和斯巴醇(3.20%)等。化香树果序挥发油对人肝癌HepG2细胞、人鼻咽癌CNE-2细胞及人宫颈癌HeLa细胞均表现出不同程度的抑制活性,IC50值分别为848.554、451.835、301.253μg/mL。结论:化香树果序挥发油具有多种活性成分,对人肝癌HepG2细胞、人鼻咽癌CNE2细胞及人宫颈癌HeLa细胞均显示出了一定的抑制活性。 Objective: To analyze the chemical constituents of the essential oil from Platycarya strobilacea and investigate the antitumor activity in vitro. Methods: The essential oil of Platycarya strobilacea was extracted by steam distillation. The chem-ical components were separated and analyzed by GC-MS. The proliferations and half inhibition concentration (IC50) were ob-served by MTY assay in hepatocellular carcinoma cell HepG2,nasopharyngeal carcinoma cell CNE2 and cervical carcinoma cell HeLa, which treated with different concentrations of The essential oil of Platycarya strobilacea. Results: There were 28 com-pounds identified and accounted for 94.10% of the peak area from the total ion-current chromatogram. The higher content of components were (Z,Z,Z)-9, 12,15-calendic acid (44.48%), hexadecanoic acid (23.30%) and barbatol (3.20%), et al. The essen-tial oil showed different antitumor activities against the proliferation of HepG2,CNE2 and HeLa cells in concentration-dependent manner. The IC50 values of The essential oil of Platycarya strobilacea against HepG2, CNE2 and HeLa cells were 848.554, 451.835,301.253 μg/mL respectively. Conclusion: The essential oil of Platycarya strobilacea contains various constituents which showed antitumor activities on HepG2, CNE2 and HeLa ceils in vitro.
