摘要
目的观察姜黄提取物对Na2S2O4诱导SK-N-SH细胞凋亡的保护作用及其机制。方法取相同代的SK-N-SH细胞随机分为正常对照组、模型组、姜黄提取物低浓度(20μmol/L)组、姜黄提取物中浓度(40μmol/L)组、姜黄提取物高浓度(80μmol/L)组。用分光光度法检测姜黄提取物对细胞外SOD、MDA的影响,通过MTT比色法检测细胞存活率,TUNEL法检测细胞凋亡率,采用Western blot检测法检测细胞Caspase-3蛋白的表达情况。结果与模型组比较,姜黄提取物各组细胞存活率升高(P<0.01),姜黄提取物各组SK-N-SH细胞培养液内的MDA水平均明显降低(P均<0.01),而SOD水平均明显升高(P均<0.01),同时Caspase-3活性受到抑制。结论姜黄提取物能够通过抗氧化作用抑制Na2S2O4对SK-N-SH细胞凋亡作用。
Objective It is to observe the protective effect of turmeric extract on SK - N - SH cells apoptosis induced by Na2S204 and to investigate the mechanism. Methods The same SK - N - SH cells were selected and randomly divided into five groups : normal control group, model group, low, moderate and high concentration of turmeric extract objects ( treated with 20 μmol/L, 40 μmol/L and 80 μmol/L respectively) groups. Cell viability was detected by MTT assay, the effects of turmeric extract on extracellular SOD, MDA were detected by Spectrophotometric method. The apoptosis rate of SK - N - SH cells was detected by TUNEL assay, and the expression of Caspase - 3 protein in the cells was determined by determined by Western blot assay. Results Compared with the model group, the survival rates of the cells increased obviously in every turmeric extract group (P 〈 0.01 ) , and the levels of MDA decreased and the levels of SOD increased obviously in SK-N-SH cells cultured in fluid(P 〈 0.01 ) , at the same time the activity of Caspase - 3 was inhibited. Conclusion Turmeric extract can inhibit the apoptosis of SK-N-SH cells induced by Na2S2O4 by its antioxidation.
