摘要
目的:通过观察大鼠脑出血后神经细胞凋亡与Bax、Bcl-2蛋白表达的动态变化及蛭龙活血通瘀胶囊的干预作用,探讨蛭龙活血通瘀胶囊可能的脑保护作用。方法:将大鼠随机分为假手术组、模型组、蛭龙活血通瘀胶囊低、中、高剂量组。采用自体血注入法制备脑出血大鼠模型。采用TUNEL法测定凋亡细胞的表达,免疫组化法测定Bax、Bcl-2蛋白的表达。结果:与模型组比较,蛭龙活血通瘀胶囊低、中、高剂量组的Bax蛋白及凋亡细胞表达明显下降,而Bcl-2蛋白的表达明显升高,差异有统计学意义(P<0.01)。与低剂量组比较,中、高剂量组各时间点的Bax蛋白及凋亡细胞表达均明显下降,而Bcl-2蛋白的表达明显升高,差异有统计学意义(P<0.01)。结论:蛭龙活血通瘀胶囊能明显促进Bcl-2表达,抑制Bax表达,减少细胞凋亡,对脑出血大鼠具有明显的脑保护作用,且存在一定量效关系。
Objective: To observe the effect of Zhilong Huoxue Tongyu capsule (ZHTC) on nerve cell apoptosis, the pro- tein expression of Bax and Bcl - 2 in intracerebral hemorrhage rats. According to those we can further elucidate ZHTC's possible effects on brain protection. Methods: The rats were randomly divided into five groups: sham-operation group, model group, low dose ZHTC group, moderate dose ZHTC group and high dose ZHTC group. Autologous blood injection method was used to produce cerebral hemorrhage model. TUNEL method was made for determining the apoptotic ceils expression and immunohisto- chemical method for determining the expression of Bax, Bcl-2 protein. Results: Compared with the model group, the apeptotic cell and the expression of Bax protein of each ZHTC group significantly decreased (P〈0. 01 ), the expression of Bcl-2 protein significantly raised (P〈0. 01 ) ; Compared with the low dose ZHTC group, the apoptotic cell and the expression of Bax protein of moderate dose ZHTC group and high dose ZHTC group significantly decreased (P〈0.01), the expression of Bcl-2 protein sig- nificantly; raised (P〈0. 01). Conclusion: ZHTC could obviously promote Bcl-2 expression, inhibit Bax expression and reduce the apoptosis. The protective effect of ZHTC for ICH rats has a certain dose-effect relationship.
