摘要
目的:通过观察多柔比星肾病大鼠核因子-KB、白介素-8水平变化,探讨肾茶联合泼尼松对肾病综合征的保护机制。方法:50只雄性Wistar大鼠随机分为正常组,模型组,肾茶组及肾茶与泼尼松联合治疗组。除正常组大鼠外单次尾静脉注射多柔比星5mg/kg来制备肾病模型,用酶联免疫吸附法检测核因子-KB、白介素-8。结果:各组大鼠尿蛋白、核因子-KB及白介素-8均明显高于正常组(P<0.01);肾茶组和联合治疗组的尿蛋白、核因子-KB及白介素-8均低于肾病组(P<0.01),但联合治疗组降低更明显,与肾茶组比较差异有显著性(P<0.01)。结论:肾茶与泼尼松联合治疗组可有效降低核因子-KB和白介素-8水平,其机制可能通过下调核因子-KB和白介素-8表达,从而对多柔比星肾病大鼠起保护作用。
Objectives:To explore the protection mechanisms spicatus and prednisone in nephrotic syndrome by observing the level of Nuclear FactorKB and Interleukin-8 of rats with doxorubicin nephropathy.Methods:50 male Wistar rats were randomly divided into normal group,model group,spicatus group and combined group of spicatus and prednisone.Except for the normal group rats a single intravenous injection of doxorubicin 5mg/kg to prepare Nephropathy model.We used ELISA to detect Nuclear FactorKB,Interleukin-8.Results:The nephrotic rats urinary protein,nuclear FactorKB and Interleukin-8 were significantly higher than those in normal group at the same period(P 0.01).In spicatus group and combined treatment group,urinary protein,Nuclear FactorKB and interleukin-8 were lower than those in ephropathy group(P 0.01),but the decline in combined treatment group was more significant,and the difference was significant compared with spicatus group(P 0.01) . Conclusions:The level of Nuclear FactorKB and Interleukin-8 in spicatus and prednisone in combined group can be effectively reduced,and the mechanism may play a protective role in the rats with doxorubicin nephropathy by decreasing the expression of Nuclear FactorKB and Interleukin-8.
