川芎嗪、黄芪合用对脑缺血再灌注后脑组织c-jun蛋白表达影响

EFFECT OF LIGUSTRAZINE AND ASTRAGUS ON EXPRESSION OF c-jun IN RAT BRAINS AFTER CEREBRAL ISCHEMIA- REPERFUSION INJURY

目的 :探讨局灶性脑缺血再灌注后脑组织c jun蛋白的表达及川芎嗪、黄芪对其表达的影响。方法 :成年雄性SD大鼠 2 4只随机分成 4组。A组 :模型组 ,即脑缺血再灌注组 (n =6) ;B组 :川芎嗪治疗组 (n =6) ;C组 :黄芪治疗组 (n =6) ;D组 :川芎嗪、黄芪合用组 (n =6)。采用免疫组化与医学图像分析系统相结合的方法 ,测定 4组大鼠大脑皮层c jun蛋白阳性细胞平均灰度值。结果 :4组大鼠脑组织缺血侧c jun蛋白阳性细胞平均灰度均低于对侧 ,即非缺血侧 (P <0 .0 1) ;在缺血侧脑组织 ,B、C、D 3组分别与A组比较 ,c jun蛋白阳性细胞平均灰度均明显升高 (P<0 .0 1) ,D组与B、C组比较 ,c jun蛋白阳性细胞平均灰度明显升高 (P <0 .0 1)。结论 :局灶性脑缺血再灌注后脑组织c jun蛋白表达显著升高 ;川芎嗪、黄芪以及两药合用均可部分抑制c jun蛋白表达 ;两药合用比单用川芎嗪或黄芪降低c jun蛋白表达的作用更显著 ,这可能是临床使用益气活血法治疗缺血性脑卒中的机制之一。

Objective:To investigate the expression of c-jun protein in rat brains after focal cerebral ischemia followed by reperfusion and effects of ligustrazine,astragus.Method:24 SD adult male rats were divided into 4 groups at random.Group A:model group or ischemia and reperfusion group(n=6);group B:treated with ligustrazine(n=6);group C:treated with astragus(n=6);group D:treated with ligustrazine and astragus(n=6).The immunohistochemistry and medical image processing system(MIPS) were used to measure mean grey level of c-jun positive cells in rat cerebral cortex of 4 groups.Result:In 4 groups,mean grey level of c-jun positive cells was lower in ischemic sides than that in non-ischemic sides(P<0.01),In ischemic sides,mean grey level of c-jun positive cellsin group B or C or D was higher than that in group A(P<0.01),mean grey level of c-jun positive cells in group D was higher than that in group B or group C(P<0.01).Conclusion:Expression of c-jun protein increased significantly after cerebral ischemia and reperfusion.Ligustrazine,astragus,ligustrazine and astragus all could inhibit partly the expression of c-jun protein.Prescription of ligustrazine and astragus has stronger inhibiting actions than ligustrazine or astragus.It may be one of mechanisms that ischemia stroke is treated by reinforcing Qi and activating blood circulation therapy in TCM clinics.
