摘要
目的 :观察增殖细胞核抗原(PCNA)在肝癌组织中的不同表达.探讨PCNA与肝癌生物学行为 之间的关系,评价PCNA可否作为一个新的客观预测肝癌发生、发展、转移及预后的可靠生物学指标。方 法:采用鼠抗人 PCNA单克隆抗体.对 83例原发性肝细胞癌. 9例胆管细胞癌. 11例转移性肝癌标本进行 了 ABC免疫组化染色,以 X2检验、 t检验、秩和检验及 log rank检验对结果做统计分析。结果:在原发性肝 细胞癌、胆管细胞癌、转移性肝癌,PCNA阳性表达率分别为87.95 %、77.77%、81.82%。PCNA阳性表达 与原发性肝细胞癌的病理分级及血清AFP明显相关,但与原发性肝细胞癌病人性别、年龄及HBsAg是否阳 性无关。结论:PCNA的表达与原发性肝细胞癌的临床病理生物学行为密切相关,PCNA有望成为预测肝癌 发生、发展的生物学指标。
Proliferating cell nuclear antigen (PCNA). also known as cyclin, is a cell cycle-related nuclear protein which accumulates in late G1 and S phases of proliferating cells. Previous researches have demonstrated it is a potentially useful tool for the study of proliferative activity and have correlated it with tumor grade. AIM: To investigate differential expressions of PCNA in liver carcinomas. and to evaluate whether PCNA can be a useful marker in predicting the occurrence. development and metastatic potential of HCC and prognosis of the patients with HCC. Methods 83 cases of hepatocellular carcinoma (HCC). 9 cases of cholangiocarcinomas (CCC). 11 cases of hepatic metastatic carcinoma (HMC). Surgically resected specimens were studied immunohistochemically by using a mAb against PCNA. An antigen retrieval method followed by a routine ABC Process was employed. Results PCNA was detected in the nucleus of cancer cells in 87. 95 %, 77. 77 %, 81. 82 % of HCC, CCC, HMC. There was a significant correlation between PCNA immunoreactivity and both histologic type of the tumor and AFP of serum, but no significant correlation between the expression of PCNA and sex. age, HBsAg. Conclusions The expression of PCNA is correlated with the occurrence, development of HCC. PCNA is hopefully to be a useful marker for the clinic of HCC.
