摘要
以脱淀粉、脱蛋白的脱脂米糠为原料,对纤维素酶水解米糠提取阿魏酰低聚糖(FOs)的工艺进行了研究。在单因素实验的基础上,以FOs的提取量为响应值,采用响应面分析法对提取条件进行优化,得到最佳提取条件为:pH4.16,反应温度41.48℃,加酶量0.29 mg/g,底物浓度3.17%,反应时间11.76 h,在此条件下,FOs的提取量达到28.48μmol/g。米糠阿魏酰低聚糖对DPPH·和·OH均有一定的清除作用。其对DPPH·、·OH的EC50分别为0.750 5 mg/mL和3.270 5 mg/mL。
In order to improve the extraction efficiency of feruloyl oligosaccharides (FOs) from defatted rice bran using cellulase hydrolysis, single factor experiments and response surface analysis were employed to optimize the enzy- molysis. The results suggested that the optimal extraction conditions were as follows: pH 4.16, reaction temperature 55℃ , enzyme addition amounts 0.29 mg/g, and reaction time 11.76 h. Under these conditions, the extraction a- mount of FOs reached 28.48 μmol/g. And the scavenging effect of FOs on the DPPH ·and ·OH; ECS0 were 0. 750 5 mg/mL and 3. 270 5 mg/mL, respectively.
