摘要
目的检测miRNA在苯并(a)芘(BaP)致癌过程中的表达变化及功能。方法利用BaP处理人支气管上皮细胞,获得了不同转化阶段的细胞模型(HBER、HBERNT、HBERT),并利用生物芯片对这些细胞中的miRNA表达进行检测,筛选可能参与BaP致癌过程的miRNA。结果在可检测到的856个miRNA中,我们发现20个miRNA与细胞转化相关,其中12个miRNA在BaP诱导细胞转化早期就有表达变化,进一步筛选发现miR-638在细胞转化时表达下调,并且可以抑制细胞的增殖和克隆形成能力,可能参与细胞转化的过程。结论 miRNA参与了BaP诱导细胞转化的过程,并发挥了重要功能,提示可作为BaP致癌早期的生物标志。
Abstract: Objective To We want to explore the expression change and function of miRNAs in the carci nogenesis of BaP. Methods We obtained cell models (including HBER, HBERNT and HBERT) at different transformation stage induced by BaP. By using microarray, we detected the expression change of miRNA pro files in the cell lines, and screened a series of miRNAs getting involved in the process of BaP induced cell trans formation. We also used cell growth curve and clone formation assays to study the function of these candidate miRNAs. Results them were found their --638 are expressed in clusion The data MiRNAs are suggest that In 856 miRNAs that we detected, 20 of them are cell--transformation related, and 12 of expression changed at the early stage of cell transformation. Further, we found that miR transformed cells and it can suppress the rate of cell growth and clone formation. Con involved in cell transformation induced by BaP and play an important role in the process. they be used as the early biomarkers of carcinogenesis induced by BaP.
