摘要 摘要:目的探讨MicroRNA--429(miR--429)在肝细胞癌(Hepatocellularcarcinoma,HCC)中的表达情况及其与HCC临床病理特征和预后的关系。方法收集来自黄曲霉毒素BI高暴露区TNM分期为l~Ⅱ期的HCC患者(138例)手术切除标本,通过TaqMan--PCR技术检测miR--429在癌组织、癌旁组织、肝癌细胞株SMMC--7721以及非肝癌细胞.株QsG-7701中的表达情况,通过风险比例回归模型及逻辑回归模型分析miR--429表达与HCC临床病理学特征以及预后的关系。结果与癌旁组织相比,miR--429在HCC癌组织中的表达水平上调,升高约4倍(P〈O.01),细胞学实验有类似发现;miR-429表达水平与HCC肿瘤大小及AFBl-DNA加合物水平相关-风险值分别为3.13(1.47~6.67)和2.70(1.28~5.56);高miR--429表达影响HCC5年期总体生存率及肿瘤无复发生存率(P〈O.01),增加总体死亡风险及肿瘤复发风险分别为3.64倍和5.94倍。结论miR--429表达参与HCC肿瘤化过程,并影响HCC预后。 Abstract: Objective To explore (HCC) and whether miR--429 having microRNA--429 (miR--429) expression in the hepatocellular carcinoma effects on the pathological features and prognosis of HCC. Methods A total of 138 patients with TNM--stage HCC from high exposure to Aflatoxin B1 (AFB1) were recrui-ted and the corresponding surgical resection samples were collected. MiR--429 expression in the tissue samples (including tumor tissues and adjacent tissues of HCC) and cells (ineluding liver cancer cell line SMMC--7721 and non--cancer liver cell line QSG--7701) were tested by using TaqMAN--PCR technique. Cox's proportion al hazard regression model and logistic regression model were used to analyze the relationship between miR-- 429 expression and HCC pathological features and prognosis. Results Compared with adjacent tissues, ex- pression level of miR--429 in tumor tissues increased by about 4 times ( P 〈0.01); similar findings was also observed in the cell analysis. Expression levels of miR--429 were correlated with primary tumor size and AFB1 --DNA adducts level, and the corresponding risk value was 3.13 (1.47-6.67) and 2.70 (1.28-5.56), re spectively. Elevated level of miR--429 expression is effective on 5--year overallsurvival rate and tumor recur rence--free survival rate (Pal0.01). The total death risk increased by 3.64 times and tumor recurrence risk increased by 5.94 times. Conclusion These results suggest that miR--429 expression be involved in HCC tumorigenesis and be associated with cancer prognosis.
