油茶组培苗生根的解剖学机理研究 被引量:7

摘要 应用光学显微技术对油茶(Camellia oleifera Abel.)组培苗茎、愈伤组织及生根过程进行石蜡切片解剖学观察,对油茶生根的机理探讨提供解剖学依据,旨在优化油茶组培生根培养条件。结果表明:油茶增殖苗开始接种到生根培养基时茎内没有根原始体,生根类型为诱导生根型。组培苗不定根根原基源于髓射线与形成层交接处的细胞分化而成。诱导根原基时间比较长,生根时间长。瓶内生根苗的愈伤组织为不规则的薄壁组织,没有观察到根原基。愈伤组织的存在,对根原基的产生有一定的刺激作用,因此,适当诱导出愈伤组织对油茶不定根的生成起到重要的作用。 Discussing the rooting mechanism of Camellia oleifera Abel with light microscopy method. Stems,callus and rooting procedure was investigated with paraffin method, designed to optimize the conditions for rooting medium. The results were as follows: there was no root primordium in multiplication seedlings. The adventitious roots of Camellia oleifera belonged to the induced root primordial. The origin of adventitious root was from the pith ray. Induced root primordial long time and rooting for a long time. The callus was irregular parenchyma with rich inclusions, and there were no root primordium in callus. The callus has stimulative effect on root primordium, so appropriately induce callus play an important role on formation of adventitious roots.
