摘要
利用优化的辣椒SRAP—PCR反应体系,研究宿州辣椒地方品种的遗传多样性.从30个引物组合中筛选出条带清晰、多态性好的15个引物组合,共扩增出62条谱带.其中,多态性谱带有40条,多态率为64.5%;不同引物扩增的条带为3~7条,谱带大小范围为100~1000bp.供试品种的遗传相似系数为0.52~1.00之间,当遗传距离D=0.52时,可将27份材料分为两大类.结果表明,宿州辣椒种质资源遗传多样性丰富.
The SRAP molecular marker was used to identify the genetic differences of pepper varieties. On the basis of orthogonally optimized SRAP-PCR amplification system, genetic diversity of 27 pepper varie- ties in Suzhou was studied. 15 effective primer combinations were selected from 30 primer-combinations. A total of 62 bands were detected with those primers and 40 of them were polymorphic. The percentage of polymorphy was 64.5 ~. Their Genetic similitary coeffient ranged from 0.52 to 1.00. Most of the am- plified fragments had a length ranged from 100 to 1000 bp. The cluster analysis using DPS software showed that the tested materials could be categorized into 2 groups at D = 0. 52, the cluster results showed that the genetic differentiation of local pepper varieties in Suzhou was abundant.
