摘要
采用高效液相色谱技术分析了纯化后的红旱莲黄酮中的主要黄酮组分,通过测定抑制率和抑制动力学,评估了红旱莲总黄酮对酪氨酸酶的抑制作用及抑制机理。结果表明,槲皮素是纯化后的红旱莲黄酮中的主要黄酮成分,其后依次为芦丁、黄芪苷、山奈酚、金丝桃苷和异槲皮素,其总黄酮含量为44.7%。与阳性对照曲酸相比,红旱莲黄酮是一种有效的酪氨酸酶抑制剂(IC50=46.99μg·mL-1),其抑制类型是典型的可逆竞争型,抑制常数Ki为34.07μg·mL-1。
The main components of purified flavonoids from Hypericum ascyron L.(HAF)were determined by high-performance liquid chromatography(HPLC).The inhibitory effect on tyrosinase and its inhibitory mechanism of HAF were evaluated through the testing of inhibition rate and inhibition kinetics.The results showed that quercetin was the major flavonoid compound of HAF,followed byrutin,astragaloside,kaempferol,hyperin and isoquercetin.The total flavonoid content was 44.7%.The tyrosinase inhibition assay indicated that the HAF exerted effective inhibitory effect against tyrosinase(IC50=46.99μg·mL-1)compared to kojic acid.The tyrosinase inhibition type of HAF was the classical pattern of reversible and competitive inhibition with inhibition constant(Ki)value of 34.07μg·mL-1.
