波棱瓜子药材指纹图谱研究 被引量:3

摘要 目的:建立波棱瓜子药材的HPLC指纹图谱,为波棱瓜子药材的质量控制提供保证.方法:甲醇提取物依次用石油醚、乙酸乙酯萃取,乙酸乙酯萃取物用甲醇溶解定容,制备样品溶液;采用RP-HPLC分析,色谱条件为WondaSil C18色谱柱(4.6mm×150mm,5μm),流动相A为50mM磷酸二氢钾缓冲盐水溶液(pH2.6),流动相B为甲醇,程序梯度洗脱,流速0.7mL/min,柱温25℃,检测波长254nm,进样量5μL.利用中药指纹图谱相似度评价系统(2004A版)对不同批次波棱瓜子药材HPLC图谱进行分析比较.结果:建立了波棱瓜子药材的HPLC指纹图谱,以Herpetolide A为参照峰确立了波棱瓜子药材指纹图谱中的18个共有峰.结论:建立的指纹图谱的重现性及稳定性均好,可用于波棱瓜子药材的质量管理规范. This paper deals with the establishment of the chromatographic fingerprint for Herba Herpetospermum caudigerum Wall by RP-HPLC by means of Methanol extract partitioned with petroleum ether and ethyl acetate successively.Fraction of ethyl acetate has then been dissolved in methanol,preparation of sample solution and analyzed by RP-HPLC with a WondaSil C18column(4.6mm ×150mm,5μm), Gradient elution with A(water containing 50mmol Potassium dihydrogen phosphate buffer salt pH=2.6)B(methanol)at 25℃,and monitored with DAD detector at 254nm.The flow rate is 0.7mL/min,and sample volume injected was 5μL.The chromatograms of different batches of Herba H.caudigerumhave been compared by the software of Similarity Evaluation System for Chromatographic Fingerprint of TCM(Version 2004A).Results show that HPLC Fingerprint of Herba H.caudigerum has been established with herpetolide A as the reference compound.Eighteen mutual peaks have been selected as the fingerprint peaks of H.caudigerum Wall.It can be concluded that the method can be provided as a scientific and effective way for quality control of H.caudigerum.
