^(125)I粒子联合黄连素治疗荷人肝癌裸鼠移植瘤的实验研究 被引量:1

摘要 目的:探讨125I粒子联合黄连素对荷人肝癌裸鼠移植瘤生长和细胞增殖及凋亡相关蛋白的影响。方法:建立人肝癌HEP-G2细胞移植瘤模型的Balb/c雌性裸鼠24只,随机分成4组,每组6只,A组为空白组,给予植入空白粒子并每天灌胃等量生理盐水;B组为黄连素组,给予植入空白粒子并按45mg/kg每天灌胃黄连素溶液;C组为125I粒子组,按治疗计划给予植入125I粒子(0.8mCi/粒)并每天灌胃等量生理盐水;D组联合治疗组,按治疗计划给予125I粒子(0.8mCi/粒)并按45mg/kg每天灌胃黄连素溶液。观察小鼠的存活情况及瘤体的生长情况,计算抑瘤率,到28d处死,制作肿瘤组织标本,进行免疫组织化学检测Ki-67表达、bcl-2/bax的比值。结果:125I粒子联合黄连素治疗组肿瘤体积最大抑制率为77.62%,与其他组比较有显著性差异(P<0.01);Ki-67表达、bcl-2/bax比值与其他各组比较具有显著性差异(P<0.01)。结论:125I粒子联合黄连素能够抑制肿瘤细胞增殖或促进肿瘤细胞凋亡。 Objective:To explore the effect and influence of ^125I seed combined with berberine on tumor growth, cell proliferation and related protein to treat human hepatocellular carcinoma (HHC). Methods:We successfully es- tablished model of liver cancer in 24 Balb/c female nude mice, and randomly divided into 4 groups ( n = 6 ) : group A for the control group, given implantation of blank particles and daily intragastric administration of NS ;group B for the berberine group, given implantation of a blank particles and 45mg/( kg · d) orally berberine solution;group C for the ^125I particle group, was given ^125I implantation of a particle (0.8mCi) according to TPS and daily gived NS;group D for ^125I particles combined with berberine group,was given 12si implantation of a particle (0.8mCi) according to TPS and 45mg/( kg · d) orally berberine solution:To observe general situation of mice, and tumor growth. 28 days later, nude mice were sacrificed and routine pathological examination was performed. The expression of proliferating cell nuclear related antigen ( Ki - 67 ), bcl - 2 and bax were detected by immunohistochemical methods. Results : The maximum tumor inhibition rate of treated group was group D, and the inhibition rate was 77.62%, there was significant difference from the other groups ( P 〈 0.01 ). The expression of Ki - 67 and the ratio of bcl - 2/bax in D group were also lower than those in the other, there was significant difference from the other groups (P 〈 0.01 ). Conclusion:^125I seed combined with berberine can effectively inhibit cell proliferation and the expression of related protein.
