氧化白藜芦醇在大鼠体内的药动学及LC-MS测定法研究 被引量:1

摘要 目的:为考察氧化白藜芦醇在大鼠体内的药动学过程,建立液质联用测定方法。方法:采用液液萃取法对大鼠血浆样品进行预处理,建立以卡马西平为内标,色谱柱为Alltima HPLC C18column(100 mm×2.1 mm,3μm),流速为0.4 mL·min-1的LC-MS/MS检测方法。结果:该方法可以用于大鼠给药氧化白藜芦醇后的血药浓度。结论:该方法耗时短(每个样品仅用3 min),回收率高,专属性强,且准确度和精密度均可满足体内药物分析的要求。 Objective: To develop and validate a sensitive and selective liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) method for determination of oxyresveratrol in rat plasma. Methods: The ana- lyre and internal standard (carbamazepine) were extracted from plasma samples using simple liquid-liquid extrac- tion technique with ethyl acetate and chromatographed on Alhima HPLC Cls column (100 mmx 2. 1 ram, 3 p.m) -1 coupled with guard column using acetonitrile-0.2% formic acid as mobile phase at a flow rate of 0.4 mL. rain The detection was carried out in multiple reaction monitoring (MRM) mode via positive electrospray ionization (ESI) interface. Results: The method was validated in terms of linearity, specificity, matrix effects, sensitivity, recovery, and accuracy. Conclusion: The method can be used for determination of oxyresveratrol in rat plasma.
