摘要
目的 建立测定肝颗粒中绿原酸含量的方法.方法 采用高效液相色谱法.色谱柱为WondaSil C18(4.6mm×150mm,5μm),流动相为乙腈-0.1%磷酸水溶液(20:80,V/V),流速为1.0mL·min-1,检测波长为327nm,柱温为25℃,进样量为20μL.结果 绿原酸的质量浓度线性范围为8.0~126.8μg·mL-1(r=0.9997,n=6);平均加样回收率为98.6%,RSD为0.66%(n=9).结论 本方法快速、简便、准确、重现性良好,可用于该制剂的质量控制.
OBJECTIVE To establish the method for the content determination of Chlorogenic acid in Gansu granules. METHODS HPLC method was adopted. Chromatographic conditions were as follows: WondaSil C18(4. 6mm × 150mm,5μm) column was used with mobile phase consisted of acetonitrile-0. 1% phosphoric acid(20: 80 ,V/V)at a flow rate of 1.0mL·min-1 under a detection wavelength at 327nm. The column temperature was 25℃, the injection volume was 20μL. RESULTS The concentration of chlorogenic acid was linear in the range of 8 - 126.8μg. mL-1 (r =0.9997,n =6) with an average recovery rate of 98.6%,RSD =0.66% (n =9) CONCLUSION A rapid, simple, accurate and reproducible method for the quality control of the preparation is established.
