摘要
对金枪鱼皮脱脂和预处理的工艺进行研究,以制备金枪鱼皮胶原蛋白肽。试验结果表明,金枪鱼皮脱脂和预处理的最佳工艺:金枪鱼皮与乙醚以13(W/V)的比例浸泡脱脂72h,乙醚溶剂换液2次后,脂肪残留率为0.12%;脱脂金枪鱼皮与0.25mol/L NaOH溶液以130(W/V)的比例在4℃下脱杂蛋白3h,得到纯度为94.87%的胶原蛋白。再将脱杂金枪鱼皮在60℃热处理15min,肽得率达27.36%。
Tuna skin was pretreated tor preparing collagen peptide by defatting and pretreatment method.The results showed that the optimal technology conditions as follows:ratio of substrate to ether 1 ∶ 3(W/ V),defatting time 72h,exchanging ether solvent 2 times,then ratio of substrate to 0.25mol/L NaOH solution 1 ∶ 30(W/V),removing non-collagen protein time 3h,temperature 4℃.Enventually,the collagen that purity reached 94.87% was heated at 60℃ for 15 min,under which the peptide yield was 27.36%.
