RyR1基因沉默对人肺癌细胞生物学活性的影响 被引量:1

摘要 目的:利用针对兰尼碱受体1(Ryanodinereceptor1,RyRl)的小干扰RNA(siRNA),研究沉默RyRl基因表达对人肺癌细胞株A549细胞生物学活性的影响。方法:(1)设计并合成针对RyRl基因的siRNA及阴性对照片段(不针对任何已知人的mRNA)和阳性对照片段(沉默GAPDH内参基因),转染肺癌细胞A549;(2)实时荧光定量一PCR(Real—timefluorogenticquanti—tativePCR,FQ-PCR)和蛋白质印迹(Westernblot)分别检测转染后RyRImRNA表达及RyRl蛋白表达的变化;(3)MTT实验检测转染前后细胞增殖能力的变化;(4)流式细胞术检测转染前后细胞周期及细胞凋亡的状态。结果:(1)FQ-PCR结果显示RyRlsiRNA-1969可下调RyRlmRNA的表达,相对表达量与阳性对照组和阴性对照组比较差异有统计学意义(P〈0.01);(2)转染RyRlsiRNA后RyRl蛋白表达下调;(3)MTT实验显示RyRl表达沉然后的A549细胞生长缓慢,与对照组相比较差异有统计学意义(P〈0.05);(4)转染后细胞凋亡率较空白对照组(正常培养的A549细胞)和阴性对照组(转染阴性对照小RNA片段的A549细胞株)显著增高(P〈0.001),处于G0~G1期的细胞明显增多(P〈0.001)。结论:RyR1基因可能为肺癌的基因治疗提供新策略。
