摘要
目的研究植入前暴露邻苯二甲酸单丁酯(DBP)对孕早期胚胎植人和发育的影响。方法在孕1.5 d、2.5d、3.5d给孕鼠灌胃300 mg·kg^(-1)·d^(-1)的DBP,然后于7.5 d、8.5 d、9.5 d统计子宫植入点数、活胎数量、吸收胎数量和胚胎长度,并使用全胚免疫荧光染色的方法检测nestin蛋白的表达。结果 DBP暴露显著降低了胚胎长度(P<0.05);降低了妊娠率(P<0.05);也降低了植入点数和胚胎数,但差异不显著;但显著提高胚胎吸收率(P<0.05)。利用神经干细胞特征性标志物nestin检测DBP对神经系统发育的影响发现,DBP处理组胚胎头部荧光强度明显低于对照组。结论孕早期DBP暴露降低了胚胎妊娠率,延缓了胚胎的发育,并降低了头部神经细胞的分化能力。
Objective: To explore the effect of preimplantation exposure of dibutyl phthalate(DBP)on embryo implantation and development in mouse. Methods: The mice were given DBP by gastric intubation at 300 mg· kg-1· d-1 on Days 1.5,2.5, 3.5 of pregnancy. The pregnancy outcome was determined on Days 7.5,8.5,9.5 of pregnancy. Number of live fetus, implantation sites, absorption and length of embryos were recorded. Expression of nestin was detected by immunofluorescence analysis. Results: DBP significantly decreased the length of fetus and pregnancy rate (P 〈 0.05), and significantly increased absorption rate of embryo(P〈0. 05). The number of fetus and implantation sites was also decreased, but not significant. Furthermore DBP treatment decreased the immunofluorescence intensity of nestin on head of 9.5 days fetus. Conclusions: Preimplantation exposure of DBP affects not only pregnancy outcome but also embryo development.
