荔枝核黄酮类化合物的体外抗腺病毒作用 被引量:16

Antiviral Activities of Flavonoids Extracted from the Core of Litchi Chinensis Against Adenovirus in vitro

摘要 目的:探讨荔枝核提取物成分黄酮类化合物(FLC)体外抑制腺病毒(AdV)增殖的作用。方法:采用细胞病变效应(CPE)、四甲基偶氮唑盐微量酶反应比色法(MTT)等方法检测不同浓度的FLC分别作用于AdV复制周期的3个阶段的抗病毒作用。结果:FLC可在病毒感染后通过抗病毒生物合成方式抑制AdV增殖,其半数细胞毒性浓度(TC50)和半数有效抑制浓度(IC50)分别为116.83mg/L,15.46mg/L,且FLC在1.25-40mg/L浓度范围内随药物浓度增加,其抗病毒作用越明显,病毒抑制率最高达84.31%,治疗指数为7.56。但研究未发现FLC具有抗病毒吸附和直接杀伤病毒的作用。结论:FLC体外可通过抗病毒生物合成的方式明显抑制AdV在人喉癌细胞(HEp-2)中的增殖。FLC有望成为一种新的有效抗腺病毒药物。 Objective.. To investigate the effect of FLC, flavornoids extracted from the core of Litchi chinensis, against adenovirus (AdV) in vitro. Methods. MTT eolorimetric assay was employed to evaluate the antiviral aetivities of the FLC. The virus-induced cytopathic effect (CPE) was scored by light microscopy, and viral inhibitory rates were calculated. Results: The median toxic concen- tration (TC50) of FLC to HEp-2 cells was 116.83 mg/L. And the AdV indueed-CPE on the cells decreased and the viral inhibitory rates increased significantly in a dose-dependent manner with FLC treatment. It was found that FLC could inhibit viral biosynthesis with the 50% inhibitory concentration (IC50) at 15.46 mg/L, and the treatment index (TI) was 7.56, but it showed no inhibition on viral adsorption to the cells and direct virucidal effect. Conclusion: It is demonstra- ted that FLC has antiviral biosynthesis activity on AdV infection, other than prevents viral ad- sorption to the cells and direct virueidal effect.
