摘要
目的探讨胰岛素样生长因子-I(insulin-like growth factor I,IGF-I)对神经前体细胞抗凋亡的影响。方法以大鼠神经前体细胞(即Ge6细胞)为研究对象,体外培养Ge6细胞,在培养基中加入含IGF-I 10ng/mL为实验组,不加IGF-I为对照组,分别继续培养2d、4d、6d,定量实时逆转录聚合酶链式反应(qRT-PCR)检测IGF-I对Ge6细胞caspase-3mRNA表达的影响。采用细胞免疫荧光染色观察各组细胞caspase-3蛋白的阳性表达率。结果 qRT-PCR结果显示,实验组Ge6细胞caspase-3mRNA的表达较对照组低,实验组培养4d(2.007±0.297)和6d(1.194±0.244)与对照组培养4d(5.530±1.027)和6d(5.371±0.513)的差异有统计学意义(P均<0.05)。细胞荧光染色表明培养2d、4d、6d实验组Ge6细胞caspase-3蛋白的阳性表达率较对照组减少,实验组培养4d(3.5%±0.2%)和6d(5.1%±0.1%)与对照组培养4d(7.0%±0.5%)和6d(7.4%±0.4%)caspase-3蛋白的阳性表达率的差异有统计学意义(P均<0.05)。结论 IGF-I对神经前体细胞具有抗凋亡作用。
Objective To investigate the effect of insulin-like growth factor I (IGF- I ) on the anti-apoptosis of neural precursor cells. Methods Rat neural precursor cells (Ge6) were used as experiment object. 10 ng/mL IGF- I were applied to cultured Ge6 differentiated 2 d,4 d,6 d and the untreated cells as control.Quantitative reverse transcriptase PCR (qRT-PCR) was used to detect the effect of the IGF- I on caspase-3 mRNA in the Ge6 of different groups and Immunofluorescent staining were applied to obverse the positive rate of caspase-3. Results qRT-PCR demonstrated that caspase-3 mRNA level of experimental group decreased compared to that of the control group, the differences between experimental group cultured for 4 d (2. 007 ± 0. 297), 6 d (1. 194 ± 0. 244) and the control group cultured for 4 d ( 5. 530± 1. 027),6 d (5. 371 ± 0. 513) were statistically significant (P〈C0. 05). Immunofluorescent staining indicated that the caspase-3 positive rate of experimental group cultured for 2 d, 4 d and 6 d were reduced compared to that of the control group,the differences between experimental group cultured for 4 d (3.5%±0.2%),6d(5.1% ± 0.1%) and the control group cultured for 4d(7.0% ± 0. 5%),6d(7.4% ± 0.4%) were statistically significant (P〈0. 05). Conclusion IGF-I plays an anti-apoptosis role on neural precursor cells.
