美洲大蠊及其不同提取物对急性免疫性肝损伤保护作用的比较 被引量:11

摘要 目的探讨美洲大蠊及其不同提取物对刀豆蛋白A(ConA)致小鼠急性免疫性肝损伤时肝脏功能的保护作用。方法将小鼠随机分为正常对照组、模型组、虫粉混悬(100、200 mg/kg)组、醇提取(100、200 mg/kg)组、水提取(100 mg、200 mg/kg)组、醇水双体取(100、200 mg/kg)组、联苯双酯200 mg/kg组。给予小鼠美洲大蠊原虫粉及其不同提取物灌胃10 d后,采用尾静脉注射ConA 20 mg/kg制备急性免疫性肝损伤模型,8 h后称取小鼠体重,眼球取血测定血清中丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)活性;处死小鼠取出肝脏和脾脏称重,计算肝脾指数;制备肝脏匀浆测定超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)活性和HE染色下肝脏病理组织学变化。结果与模型组比较,给药组均有降低ConA致小鼠急性免疫性肝损伤肝脾指数(P<0.01,P<0.05);降低血清中ALT、AST活性(P<0.01,P<0.05)和肝脏组织中MDA的活性(P<0.01,P<0.05);升高肝脏组织中的SOD、GSH的活性(P<0.01,P<0.05);减轻小鼠肝脏病理损伤。结论美洲大蠊及其提取物对ConA引起的小鼠急性免疫性肝损伤具有一定保护作用。美洲大蠊及其提取物存在药效差别,虫粉混悬组及水提取组作用优于其醇提取组和醇水双提取组。 Objective To investigate the effects of Periplaneta American and its different extracts on acute immunological liver injury caused by coneanavalin A(ConA) in mice. Methods The model of mice acute immunological liver injury was induced by ConA on day 10 after pretreatment with Periplaneta American and its different extracts. After 8 h, the mice were weighted, and blood were collected for determinate serum alanine aminotIansferas (ALT) and aspartate aminotransferas(AST). Then the mice were killed, and the liver and spleen were weighted immediately, and the liver and spleen index were calculated. The liver homogenates were prepared to test malondiadehyde (MDA),superoxidedismutase(SOD), glutathione (GSH) and hepatic pathological examination was observed with HE coloration. Results Periplaneta American and its different extracts can decrease liver and spleen index (P 〈 0.05 ,P 〈 0. 01 ) and the activity of ALT, AST(P 〈 0. 05, P 〈 0. 01 ) in serum, and MDA ( P 〈 0.05, P 〈 0. 01 ) in liver. Moreover, it could enhance the SOD and GSH activity(P 〈 0.05 ,P 〈 0.01 )in liver tissues and thus relieved liver pathological injury, the efficacy of Periplaneta American and its different extracts existed differences. Conclu- sion Periplaneta American and its extracts have protective effects on acute immunological liver injury in mice, and the protozoa and water extracts better than alcohol extracts or alcohol-water double extracts.
