姜黄素联合顺铂对人肝癌细胞HepG2增殖、自噬和凋亡的影响 被引量:11

Effects of curcumin combined with cisplatin on cell proliferation,autophagy and apoptosis in vitro hepatocellular carcinoma HepG2 cells

摘要 目的探讨姜黄素联合顺铂对人肝癌细胞HepG2增殖、自噬和凋亡的影响。方法将不同浓度的姜黄素、顺铂单独或联合处理体外培养的人肝癌细胞HepG2。24h后,MTT法检测细胞活性,免疫荧光观察细胞内自噬泡形成情况,Western blot检测自噬、凋亡相关蛋白表达。结果与姜黄素或顺铂单药给药组相比,姜黄素联合顺铂更能明显抑制HepG2细胞的增殖(P<0.05),促进微管相关蛋白LC3的点状分布,提高自噬相关蛋白Beclin1的表达和LC3Ⅱ/Ⅰ的比值(P<0.05),降低凋亡相关蛋白Bcl-2/Bax的比值(P<0.05),并激活半胱天冬氨酸蛋白酶3(P<0.05)。结论姜黄素联合顺铂可诱导HepG2细胞的自噬和凋亡,显著提高HepG2细胞对顺铂的敏感性。 Objective To investigate the effects of curcumin combined with cisplatin on cell proliferation, autophagy and apoptosis in vitro hepatocellular carcinoma HepG2 cells. Methods HepG2 cells were treated by different concentrations of curcumin, cisplatin alone or in combination for 24 hours. Cell proliferation was detected by MTT assay. The formation of autophagic vacuoles was observed by immunofluorescence. The expressions of autophagy- and apoptosis-related protein were detected by Western blot. Results Compared with using curcumin or cisplatin alone, curcumin combined with cisplatin had more abilities of depressing cell proliferation (P〈0. 05), promoting punctate distribution of the microtubule-associated protein, increasing the autophagy related proteins Beclinl expression and the ratio of LC3 Ⅱ to LC3 T , reducing the ratio of Bcl-2 to Bax(P〈0. 05), and activating Caspase 3. Conclusion Curcumin combined with cisplatin can effectively induce autophagy and apoptosis on HepG2 cells and increase the sensitivity of HepG2 cells to cisplatin.
