摘要
目的探讨X染色体连锁凋亡抑制蛋白(XIAP)相关因子1(XAF1)在肺鳞癌发生、发展中的作用。方法 51例肺鳞癌患者术后切除的肺组织标本分为三组:A组为肺癌组织,B组为肺癌旁2cm组织,C组为肺癌旁5cm组织。应用免疫组化检测三组XAF1蛋白表达;半定量逆转录-PCR检测51例肺癌组织中XAF1和XIAP mRNA表达,分析其表达与临床病理特征的相关性。结果 A组XAF1的表达强度显著低于B、C组(54.9%vs.78.4%、86.7%)(P<0.05);低分化和高TNM分期的XAF1表达低于高分化和低TNM分期(P<0.05)。A组中XAF1mRNA的表达水平低于C组(P<0.05)。结论 XAF1在肺癌发生、发展中具有促凋亡作用。
Objective To investigate the role of tumor-suppressing gene X chromosome-linked inhibitor of apoptosis (XIAP)-associated factor 1 (XAF1) in the occurrence and development of squamous cell lung cancer. Methods With immunohistochemical method, the expressions of XAF1 protein were detected in 51 cases of squamous cell lung cancer specimens(group A) and in the tumor- adjacent tissues 2 cm(group B) and 5 cm(group C) away from cancer tissues. The expressions of XAF1 mRNA and XIAP mRNA were detected by RT-PCR in group A. Results The expression of XAF1 was lower in group A than that in groups of B and C(54. 9% vs. 78. 4% and 86.7%) (P〈0. 05), which was lower in the cancer with low differentiation and high TNM grade than that in those with high differentiation and low TNM grade (P〈0. 05). The XAF1 mRNA expression was lower in group A than that in group C(P〈0. 05). Conclusion XAF1 plays an important role in inducing apoptosis in the occurrence and development of squamous cell lung cancer.
