摘要 目的:研究塞来昔布与茶多酚合用对三阴乳腺癌MDA-MB-231细胞增殖及凋亡的影响。方法:MTT比色法检测塞来昔布、茶多酚单药及两者联合用药对乳腺癌MDA-MB-231细胞增殖的影响。Hoechst33258荧光染色观察细胞形态学改变。流式细胞术AnnexinV-FITC/PI双染检测细胞凋亡率。结果:MTT结果显示塞来昔布(10μmol/L)与不同浓度茶多酚(6.25、12.5、25、50、100μg/mL)单用及合用均对MDA-MB-231细胞产生增殖抑制作用,且联合用药组的抑制率高于各单用组,差异具有统计学意义(P<0.05)。Hoechst33258荧光染色结果显示塞来昔布(10μmol/L)与茶多酚(50、100μg/mL)合用组的凋亡细胞较单用组明显增加,出现典型凋亡形态学特征。流式细胞术进一步证明两药合用使凋亡率显著增加。结论:塞来昔布与茶多酚合用具有显著抑制三阴乳腺癌MDA-MB-231细胞增殖,诱导细胞凋亡的作用。 Objective: To investigate the effects of celecoxib combined with tea polyphenols on the proliferation and apoptosis of triplenegative breast cancer cell MDA-MB-231. Methods: MTT assay was adopted to detect the effects of celecoxib, tea polyphenols and celecoxib combined with tea polyphenols on the proliferation of breast cancer cell MDA-MB-231. Hoechst33258 staining was used to observe the morphologic changes. Cell apoptosis was detected by AnnexinV-FITC/PI double staining. Results: MTT assay showed that celecoxib (10 umol/L) and tea polyphenols (6.25,12.5,25,50,100 ug/mL) alone and in combination inhibited the proliferation of MDA-MB- 231 cells. The inhibition rate of the combination groups was higher than single drug groups and the difference was statistically significant (P〈0.05). Hoechst33258 staining showed that the number of apoptotic cells in combination groups composed by celecoxib (10 umol/L) and tea polyphenols (50,100 ug/mL) significantly increased compared with single drug groups and the typical morphological characteristics of apoptotis could be observed. Flow cytometry was further evidence that the apoptosis rate significantly increased by celecoxib and tea polyphenols combined. Conclusion: Celecoxib in combination with tea polyphenols can significantly inhibit the proliferation and induce apoptosis of triple-negative breast cancer cell MDA-MB-231.
