来氟米特对减体积性肝移植后肝再生及功能的影响

EFFECTS OF LEFLUNOMIDE ON LIVER REGENERATION AND LIVER FUNCTION AFTER REDUCE-SIZED LIVER TRANSPLANTATION IN RATS

摘要 目的 研究新型免疫抑制剂来氟米特对大鼠减体积性肝移植后肝再生及功能的影响。方法 动物分 A,B,C3组 ,来氟米特的剂量分别为 0、2 .5、5 .0 mg·kg- 1· d- 1。动物于术后 1、3、5、7d处死。每克肝组织中蛋白质含量 ,血清 AL T及总胆红素水平 ,肝细胞分裂指数 ,肝细胞增殖细胞核抗原标记指数作为肝再生的指标。结果 B组术后 3d的每克肝组织蛋白质含量 ,肝细胞分裂指数 ,肝细胞增殖细胞核抗原标记指数均高于 A,C组 (P<0 .0 5 )。A、B、C3组 AL T,总胆红素水平无显著差异。结论  2 .5 mg·kg- 1 · d- 1 来氟米特能促进大鼠减体积性肝移植后肝再生 ,对肝功能无负面影响。 Objective To study the effects of Leflunomide(Lef),a novel immunoinhibitor,on liver regeneration and liver function after reduce sized liver transplantation(RSLT) in rats.Methods Animals were devided into three groups(A,B,C).The dosage of Lef were 0,2.5,5.0 mg·kg -1 ·d -1 respectively.Animals were sacrificed at various time points(1,3,5,7d postoperatively).Protein content,serum ALT levers and total bilirubin levers,Mitotic indes(MI) of hepatocytes and proliferating cell nuclear antigen(PCNA)Labeling index(LI) of hepatocytes were used as parameters of liver regenerative activity.Results Protein content,MI and PCNA LI were higher in group B than that in group A or in group C (P<0.05).There were no significant differences in three groups about serum ALT levers and total bilirubin levers.Conclusion Lef with the dosage of 2.5mg·kg -1 ·d -1 enhances liver regeneration after RSLT in rats and has no nagitive effect on liver function.
