摘要
以灯心草为研究对象,用多糖提取率作为衡量提取工艺的指标,在单因素实验基础上,根据星点设计原理,选取超声时间、浸提温度、浸提时间、料液比四因素五水平进行响应面分析,建立灯心草多糖提取率的二次回归方程,得到最佳提取工艺.结果表明浸提时间对灯芯草多糖的提取率影响最为显著.当工艺条件为超声时间25.9 min、浸提温度78.1℃、浸提时间2.05 h、液料比86.6 1 mL/g时,灯心草多糖理论提取率为0.607 4%,验证值为0.613 2%.
The extraction technology of polysaccharide from Juncus effuses was optimized with the extraction rate of polysaccharides as the evaluation index. On the basis of sing]e test, response surface analysis was studied through the central composite design. In the optimization, a quadratic regression model predicting polysaccharides yield as a function of 4 extraction factors, i. e. , ultrasonic time, extraction temperature, extraction time and liquid-to-solid ratio was set up and the effects of the four extraction factors on polysaccharides yield were evaluated. Experiment result showed that extration time had the most significant effect on polysaccharides yield. The optimum extraction technology was determined as follows:ultrasonic time 25.9 min, extraction temperature 78.1 ~C, extraction time 2.05 h and liquid-to-solid ratio 86.6 : 1 mL/g for an extraction. Under such conditions, the predicted and experimental values of polysaccharide from Juncus effuses yield were 0.607 4% and 0. 613 2% ,respectively.
