摘要 研究肝细胞对星状细胞凋亡的影响和中药复方861的干预作用。方法:体外研究采用分离培养的正常大鼠原代肝细胞和星状细胞系,将培养的肝细胞和复方861作用于不同的培养基再作用于星状细胞,用电镜观察和流式细胞仪检测细胞凋亡。临床研究对象是用复方861治疗6个月行治疗前后肝穿的慢性乙型肝炎病人。结果:原代培养的正常肝细胞促进了星状细胞的凋亡(P<0.05)。复方861可以增加这种促凋亡作用(P<0.05)。临床研究显示慢性乙型肝炎患者肝星状细胞大量活化增生,用复方861治疗6个月后有肝细胞再生,同时活化的星状细胞数量显著减少,可观察到其凋亡。结论:正常肝细胞可以促进星状细胞凋亡,复方861对星状细胞有直接和间接的促凋亡作用。 To investigate the effect of normal hepatocytes and herbal compound 861 (Cpd 861) on apoptosis of hepatic stellate cells (HSC). Methods: The study was carried out on the primary culture of normal rat hepatocytes and hepatic stellate cell line. Cell apoptosis was detected by electron microscopy and flow cytometry. The in vivo study based on 42 patients with chronic hepatitis B, Cpd 861 was given orally three times a day to these patients at least 6 months. Liver biopsies were performed just before and after the treatment. Results: HSC apoptosis rate was increased by replacement of undiluted culture conditioned medium from normal hepatocytes in vitro (P<0. 05). Furthermore, this increase effect could be enhanced by Cpd 861 (P<0. 05). The clinical study elucidated that the HSCs were activated and proliferated in patients with chronic hepatitis B. After Cpd 861 treatment the number of activated HSC decreased and the apoptosis of HSC could be seen in the liver biopsy. Conclusion; Normal hepatocytes increased HSC apoptosis. Cpd 861 had both direct and indirect effect through hepatocytes on stimulating apoptosis of hepatic stellate cells.
