海地瓜岩藻糖基化海参硫酸软骨素对Ⅱ型糖尿病小鼠胰岛素抵抗改善作用的研究 被引量:6

摘要 目的研究海地瓜岩藻糖基化海参硫酸软骨素(fucosylated chondroitin sulfate from the sea cucumber Acaudina molpadioides,Am-CHS)对Ⅱ型糖尿病小鼠胰岛素抵抗的改善作用。方法以高脂高糖饲料饲喂法建立Ⅱ型糖尿病小鼠模型。雄性C57BL/6小鼠随机分为正常对照组、模型对照组、阳性对照组、实验A、B、C组。正常对照组饲喂标准饲料,其它组饲喂高脂饲料。阳性对照组、实验A、B、C组分别在饲料中添加罗格列酮(rosiglitazone,RSG,1mg·kg-1·d-1)、高剂量Am-CHS(80mg·kg-1·d-1)、低剂量Am-CHS+RSG(20+1mg·kg-1·d-1)、高剂量Am-CHS+RSG(80+1mg·kg-1·d-1)。各组小鼠自由摄食摄水19周。实验结束后,称重小鼠白色脂肪重量,检测空腹血糖、空腹血清胰岛素及血清脂联素、抵抗素、瘦素、肿瘤坏死因子-α(TNF-α)水平。结果 Am-CHS可显著降低Ⅱ型糖尿病小鼠的脂肪积累,降低血糖和胰岛素水平,改善胰岛素抵抗,提高血清脂联素含量,降低抵抗素、瘦素和TNF-α含量。Am-CHS与RSG复配使用,效果更显著。结论 Am-CHS能显著改善Ⅱ型糖尿病小鼠的胰岛素抵抗程度,其作用机制可能与改善肥胖引起的脂肪细胞因子的分泌紊乱有关。 Objective To unravel the effects of fucosylated chondroitin sulfate from Acaudina mo/padioides (Arn-CHS) on ameliorating insulin resistance in type Ⅱ diabetic mice. Methods The model mice were established by feeding a diet of high-fat and high-sucrose. Male C57BL/6 mice were randomly assigned into the normal control group, the model control group, the positive control group, experimental A, B, and C groups. The mice in the normal control group were fed the standard diet, and the animals in the other groups were fed the high-fat diet. Rosiglitazone (RSG, 1 mg kg^-1 d^-1), high-dose Am-CHS (80 mg kg^-1 d^-1), low-dose Am-CHS + RSG (20 + 1 mg kg^-1 d^-1), and high-dose AmCHS Jr RSG (80 + 1 mg· kg^-1·d^-1) were added into the diets of the positive control group, experimental A, B, and C groups, respectively. All animals were fed with diet and water freely for 19 weeks. After experiments, the white fat weight, fasting serum glucose, fasting serum insulin, serum adiponectin, resistin, leptin, and tumor necrosis factor-α (TNF-α) levels were determined. Results Am-CHS markedly decreased fat deposition, blood sugar and insulin levels, ameliorated insulin resistance, in- creased serum adiponectin concentration, and lowered serum resistin, leptin, TNF-a levels. The effects were more significant when Am-CHS compounded with RSG. Conclusion Am-CHS significantly ameliorated insulin resistance in type Ⅱ diabetes mellitus mice, and the mechanisms might involve the regulation of adipocytokines paraseeretion caused by adiposity.
