摘要
为了研究单胺类神经递质的检测方法,试验采用RP-HPLC测定山羊海马中4种单胺类神经递质,色谱条件为Symmetry C18(4.6×150 mm,5μm)色谱柱,以甲醇与去离子水(含40 mmol/L醋酸钠、30 mmol/L柠檬酸、0.2 mmol/L EDTA-2Na、0.4 mmol/L辛烷磺酸钠)为流动相,采用等度洗脱,然后进行荧光检测。结果表明:在15 min内,4种单胺类神经递质分离完全,在2~500 ng/mL范围内线性关系良好,相关系数分别在0.99以上;变异系数在2.7%~3.8%之间;平均回收率在90%以上。说明该方法的稳定性、重现性良好。
To study a method for detecting monoamine neurotransmitter, four monoamine neurotransmitters were detected in goat hippocampus by reversed - phase high - performance liquid chromatography ( RP - HPLC) method. The ehromatogram condition were as follows : Symmetry C18 column (4.6 x 150 mm ,5 p^m) ,methanol and deionized water ( including 40 mmol/L of sodium acetate,30 mmol/L of citric acid,0.2 mmol/L of EDTA-2Na,and 0.4 mmoL/L of sodium oetanesulfonate) used as the mobile phase. The fluorescence detection was used after isocratic elu- tion. The results showed that the four monoamine neurotransmitters were separate completely within 15 min; the linear relation was good in the range from 2 to 500 ng/mL, and the correlation coefficients were above 0.99, respectively; the coefficients of variation were between 2.7 % and 3.8% ; the average recoveries were above 90%. The results indicate that the RP - HPLC method has the advantages of good stability and repro- ducibility.
