摘要
以MRS为基础培养基,对培养基的碳氮源和培养条件进行了优化,确定了植物乳杆菌C88优化培养基的配方为:果糖10g/L,葡萄糖20g/L,大豆蛋白胨26.66g/L,酵母浸粉13.33g/L,柠檬酸钠5g/L,无水乙酸钠5g/L,KMP042g/L,MgS04O2g/L,MnSO40.05g/L,吐温801.0mL/L。另外对植物乳杆菌C88高密度培养条件进行了优化,结果表明,植物乳杆菌C88在35℃条件下,同时流加20%的Na2CO3使发酵液pH值保持6.0-6.5,静止培养16h后,活菌数可达9.3×10^10mL^-1,本研究为植物乳杆菌C88冻干直投式发酵剂的制备奠定了基础。
Based on MR.S broth, the optimization medium composition and culture conditions for Lactobadllus plantarum C88 were studied. The optimized medium was obtained with fructose 10 g/L, glucose 20 g/L, soy peptone 26.66 g/L, yeast extract 13.33 g/L, sodium citrate 5 g/L, sodium acetate anhydrous 5 g/L, dipotassium phosphate 2 g/L, magnesium sulfate 0.2 g/L, manganese sulfate 0.05 g/L, tween 80 1 mL/L. The fermentation conditions were optimized using this enrichment medium, and the results showed that the viable count of lacto-bacillus plantarum C88 reached 9.3×10^10 CFU/mL under the conditions that the pH of medium was 6.0-6.5 adjusted with 20% Na2CO3, and cultivated statically at 35℃ for 16 h.
