TGF-β1、Smad3及Smad7在宫腔粘连患者粘连组织中的表达及意义 被引量:24

Expression and significance of TGF-β_1,Smad3 and Smad7 in the fibrosis tissue of patients with intrauterine adhesions

摘要 目的检测TGF-β1及Smad3、Smad7在宫腔粘连(IUA)患者粘连组织中的表达,探讨其在宫腔粘连发生中的意义。方法将30例宫腔粘连的患者作为研究组,在宫腔镜直视下取粘连组织。将继发不孕行宫腔镜检而子宫内膜正常的30例患者作为对照组,在宫腔镜直视下取正常子宫内膜。应用免疫组织化学法检测TGF-β1、Smad3及Smad7在以上组织中的表达。结果①TGF-β1和Smad3在研究组中的表达高于对照组,而Smad7的表达低于对照组,比较差异有统计学意义(P<0.05)。②研究组中TGF-β1与Smad3的表达呈正相关(P<0.05),而与Smad7的表达呈负相关(P<0.05)。结论①宫腔粘连组织中TGF-β1、Smad3及Smad7的异常表达可能与IUA的发生有关。②在TGF-β1/Smad信号通路中Smad3正调控过度和Smad7负调控不足可能促进了IUA的形成。 [Objectives] To investigate the expression of transforming growth factor-β1, (TGF-β1), Smad3 and Smad7 in the fibrosis tissue of patients with intrauterine adhesions (IUA), and to discuss the significance of these factors in the forming of IUA. [Methods] The study group was composed of 30 patients with IUA and the control group included 30 infertility patients without intrauterine adhesions. The fibrosis tissue from the study group and normal endometrium tissue from the control group were obtained at hysteroseopy, respec tively. Immunohistoehemical method (streptavidin peroxidase conjugated method) was used to determine the ex pression of TGF-β1, Smad3 and Smad7 protein in these tissues. [Results] The expression of TGF-β1, Smad3 was higher in the study group than that in the control group. (P〈0.05), but the expression of Smad7 was low er (P 〈0.05). The expression of TGF-β1 was positively correlated with that of Smad3 in the study group (P 〈 0.05), but negatively correlated with that of Smad7 (P 〈0.05). [ Conclusion] The abnormal expression of TGF- β1, Smad3 and Smad7 in the fibrosis tissue may be related to the occurrence of IUA. In the TGF-β1/Smad signaling transduction pathway, the excessive positive feedback of Smad3 and insufficient negative feedback of Smad7 may be related to the occurrence of IUA.
