摘要
目的建立成年大鼠心肌细胞缺氧/复氧损伤模型,观察吡那地尔(Pinacidil,P)后处理对心肌细胞超微结构的影响。方法体外培养成年大鼠心肌细胞,建立心肌细胞缺氧/复氧损伤模型,将细胞随机分为6组,正常组(N)、缺氧/复氧组(H/R)、缺氧后处理组(HPO)和吡那地尔不同浓度组(25μmol/L、50μmol/L、100μmol/L,P25、P50、P100组),采用透射显微镜观察各组心肌细胞缺氧/复氧损伤后心肌细胞超微结构的变化。结果透射显微镜显示N组超微结构正常,H/R组超微结构损伤严重,HPO、P25、P50、P100组损伤较轻,P50组较P25、P100组损伤更轻。结论缺氧/复氧能使成年大鼠心肌细胞造成损伤,吡那地尔后处理对此损伤具有一定保护作用。
Objective To establish the hypoxia/reoxygenation injury model of adult rats' myocardial cells and to observe the influence of Pinacidil postconditioning on myocardial ceils' uhrastructure . Methods Adult rats' myocardial cells were cultured in vitro and hypoxia/reoxygenation injury model of myocardial cells were established. Then, the cultured cells were randomly divided into 6 groups, which were normal group ( N ), hypoxia/reoxygenation group ( H/R ), hypoxia post - processing group (HPO), and group of Pinacidil with different levels of concentration (25 μmol/L (25), 50 μmol/L (P50), 100 μmol/L (P100). Finally, the uhrastructural changes of myocardial cells in each group af- ter hypoxia/reoxygenation injury could be observed by a transmission electron microscope. Results The ultrastructure of N group was presented normal by the transmission electron microscope; the uhrastruc-ture of H/R group was seriously injured ; uhrastructures of HPO group and P25, P50, and P100 group were slightly injured, among which, the ultrastructure of P50 group was less injured than that of P25 and P100 group. Conclusion H/R(hypoxia/reoxygenation) can cause injury to the adult rats' myocar-dial ceils, and Pinaeidil postprocessing has certain protective effect on this injury.
