Determination of chlorogenic acid and macranthoidin B in Flos Lonicerae from Suiyang by HPLC- ELSD method

摘要 目的 采用HPLC-ELSD联用的方法对绥阳山银花中绿原酸和灰毡毛忍冬皂苷乙进行含量测定.方法 采用Phenomenex Luna C1s(250 mm×4.6 mm,5 μm)色谱柱,柱温30℃,以乙腈为流动相A,0.4%醋酸溶液为流动相B,梯度洗脱(0~10 min,11.5% A→15%A;10~12min,15%A→29%A;12~18 min,29% A→33% A; 18~30min,33% A→45%A),流速为0.8 mL/min,蒸发光散射检测器漂移管温度40℃.结果 绿原酸和灰毡毛忍冬皂苷乙的回归方程依次是y=4 609x-300.53(R2=0.9995)和y=5 810.1x-421.19(R2 =0.998 8),其线性范围依次是0.0 804~ 0.6 432 mg/mL,0.0966~0.7728 mg/mL.平均加样回收率依次为100.5%,101.6%.结论 该方法操作简单,结果准确,绥阳山银花中绿原酸与灰毡毛忍冬皂苷乙的平均含量分别为4.45%、7.94%,均高于2010版《中国药典》的规定2.0%、5.0%,具有潜在的开发价值. Objective To determine chlorogenic acid and macranthoidin B in Flos Lonicerae from ang by HPLC - ELSD method. Methods Shimadzu High Performance Liquid Chromatography was Suiy- used, the column was Phenomenex Luna Ci8 (4.5 × 250 mm, 5 μm), and the column temperature was 30 ℃. The mobile phase was composed of acetonitrile (A)-0.4% acetic acid (B) solution with gradient elution (0 - 10 min, 11.5% A→d5% A ; 10 - 12 min, 15% A →29% A ; 12 - 18 min, 29% A→33% A; 18-30 min, 33%A→45%A), at a flow rate of 0.8 mL/min, and the drift tube temperature was 40 ℃. Results The regression equation of ehlorogenie acid and macranthoidin B were y = 4609x - 300.53 ( RE =0. 9995), y = 5810. 1x - 421.19 ( R^2 = 0. 9988 ), respectively. The linear ranges of chlorogenic acid and macranthoidin B were 0.0804 - 0. 6432 mg/mL,0. 0966 - 0. 7728 mg/mL, respectively. Their average recoveries ( n =9) were 100.5% and 101.6%, respectively. Conclusion HPLC - ELSD meth-od is simple, accurate for determining the contents of chlorogenic acid and macranthoidin B in Flos Lonicerae . The average contents of chlorogenic acid and macranthoidin B are 4.45% and 7.94% , re-spective, which are higher than those in Chinese Pharmacopeia (2010 edition). The study suggests that Flos Lonicerae from Suiyang is deserved to be further developed.
