摘要
真菌多糖是指10个以上单糖通过糖苷键连接形成直链或支链的一类糖,具有提高免疫,抗氧化,抗辐射,抗肿瘤等多种生理活性.朱红硫磺菌(Laetiporus sulphureus(Fr.)Murrill var.miniatus(Jungh.)Imaz.)是珍贵的药膳兼用菌.本研究从朱红硫磺菌子实体中,经过除杂、粉碎、热水浸提、醇沉处理后,得到了朱红硫磺菌杂聚糖粗提物,并运用DEAE-cellulose column进行柱层析分离纯化,得到朱红硫磺菌多糖纯品(LSF-A).通过硫酸苯酚法检测,水解,单糖分析,扫描电镜(SEM),红外光谱技术(IR)和核磁共振技术(NMR)对朱红硫磺菌多糖LSF-A进行了初步的结构分析.结果显示,朱红硫磺菌多糖由两种单糖构成,其结构中含有葡萄糖成分.在体外研究了不同浓度的朱红硫磺菌多糖对巨噬细胞吞噬作用的影响.结果显示朱红硫磺菌多糖具有显著的刺激巨噬细胞吞噬的作用,并且呈剂量依赖关系.这些结果说明朱红硫磺菌多糖(LSF-A)具有显著的免疫调节活性,可以作为一种理想的天然免疫调节剂资源.
Fungal polysaccharides are long carbohydrate molecules, of repeating units joined together by glycosidic bonds. They are often linear, but may also be highly branched. In recent years, more and more fungal polysaccha- rides have been reported to exhibit a variety of biological activities, including anti-tumor, immune-stimulation, an- ti-oxidation etc. A polysaccharides was extracted from the fruiting bodies of Laetiporus sulphureus (Fr.) Murrill var. miniatus (Jungh.) Imaz. eus (Fr.) with water and then successively purified by DEAE-cellulose 52 column chromatography, yielding one major polysaccharides fraction: LSF-A. Structural features of LSF-A were investiga- ted by a combination of total hydrolysis, scanning electron microscope (SEM) , infrared (IR) spectra and nuclear magnetic resonance (NMR) spectroscopy studies. The results showed that the polysaccharides LSF-A was com- posed of two monosaccharides, and one component was the glucose. The immune activity analyses revealed that LSF-A possessed considerable immune activity. Accordingly, Laetiporus sulphureus ( Fr. ) Murrill var. miniatus (Jungh.) Imaz. eus (Fr.) might serve as an effective healthcare food and source of natural immune-modulators.
