摘要
本试验建立了一种用超高效液相色谱串联质谱仪(UPLC—MS/MS)测定猪尿液中喹乙醇代谢物3-甲基喹(?)啉-2羧酸(MQCA)的残留量的检测方法。猪尿液经氢氧化钠溶液碱性条件下水解,在酸性条件下经混合型阴离子固相萃取柱净化,过0.22μm滤膜后,超高效液相色谱串联质谱仪进行定量测定。试验结果表明,MQCA浓度在5 ng/mL^100 ng/mL范围内呈良好的线性关系。当猪尿液中MQCA含量在1 ng/mL^100 ng/mL时,平均添加回收率为70%~100%之间,最低检测限为1 ng/nL。本方法操作简便、快速,结果稳定、可靠,可满足猪尿液中MQCA的含量检测。
This paper presents a method using Ultra Performance liquid chromatography-tandem mass spectrometry (UPLC- MS / MS ) to determine olaquindox metabolite 3-methyl quinoxaline-2 carboxylic acid ( MQCA ) residue in pig urine. Pig urine un- derwent alkaline hydrolysis by sodium hydroxide solution and was purified in acidic conditions by mixing anionic solid phase extrac- tion column after being tittered by a 0.22 ~m filter, it was quantitatives determined by ultra performance liquid chromatography tan- dem mass spectrometer. The test results show that, MQCA concentration in the range of 5~100 ng / mL showed a good linear rela- tionship,When the MQCA content in pig urine was at l^100 ng / mL, the average recovery was between 70% ~100%, with the lowest detection limit of 1 ng / mL. This method is simple, rapid, stable, reliable and can meet the needs of pig urine MQCA deter- mination.
